摘要
目的:建立人血浆中尼莫地平浓度的气相色谱测定方法。方法:以氢氧化钠碱化样品后用正己烷-乙酸乙酯(1:1,v/v)提取,以硝苯地平作为内标,经SE-30填充柱分离,进入氢火焰离子化检测器检测。结果:尼莫地平浓度在1~300ng/mL范围呈线性相关。最低检测限约为1 ng/mL。三种浓度的平均回收率为88.1%~95.2%,日内RSD为1.6%~7.1%,日间RSD为3.6%~5.0%。结论:本法简便、快速、重现性好,适合用于尼莫地平的人体药代动力学研究。
Objective To establish a GC method for determination of nimodipine in human plasma.Methods After adjustment to a basic pH with sodium hydroxide solution,plasma samples were extracted with n-hexane-ethyl acetate(1:1,v/v) and separated on a SE-30 column with FID as the detector.Results The linear range was 1~300 ng/mL,the detective limit was 1 ng/mL.The recovery was 88.1%~95.2% with the intra-day RSD from 1.6 to 7.1%,and the inter-day RSD from 3.6 to 5.0% at three different concentrations.Conclusion The method was simple,rapid,and successfully applied to study the pharmacokinetics of nimodipine in healthy Chinese volunteers.
