摘要
目的:探讨全氟辛烷磺酸(PFOS)对大鼠的肾脏毒性。方法:清洁级SD大鼠24只,随机等分为4组,分别用含PFOS 0mg/kg、5mg/kg、25 mg/kg、125mg/kgPFOS的饲料喂养60d后,摘取眼球收集血液分离血清,检测血清中UREA、CRE、UA的含量;分离肾脏,测定肾脏器系数;然后一侧肾作常规病理学观察,另一侧肾组织匀浆分离上清液,测定MDA、GSH含量及GSH-Px、SOD活力。结果:与对照组比较,PFOS中、高剂量组肾脏器系数增加、血清UREA、CRE、UA含量升高(P<0.05);中、高剂量组肾组织出现明显的管型和蛋白物质渗出;低、中、高剂量组肾组织中MDA、GSH含量升高(P<0.05),GSH-Px、SOD活力降低(P<0.05),呈剂量-效应关系。结论:PFOS可对大鼠的肾脏产生损伤。
Objective To study the renal toxicity of the perfluorooctane sulfonate(PFOS) in rats.Method 24 healthy SD rats were averagely divided into 4 groups randomly including control group,low dose,middle dose and high dose PFOS group,there is 6 rats in each group.The four groups were respectively fed fodder which added different dose PFOS(0,5,25 and 125mg/kg fodder).After 60 days fed,their renal organ coefficients was computed and the renal morphous was observed,the level of UREA,CRE,UA,MDA,SOD,GSH and GSH-Px were detected in rat’s nephridial tissue.Results Compared the control with middle and high dose PFOS group, rats’ renal organ coefficient and the level of UREA、CRE、UA in blood serum all rised.Rat’s nephridial tissue had obvious cast and protein exudation in middle and high dose PFOS group.The level of of MDA,GSH of nephridial tissue was increased and the activity of GSH-Px,SOD was dropped in all PFOS group,and there were dosedependent manner(P<0.05).Conclusions PFOS has renal toxicity in rats.
