褐藻糖胶联合雷帕霉素对多发性骨髓瘤细胞的增殖抑制作用

摘要 目的探讨褐藻糖胶(Fucoidan)联合雷帕霉素(Rapamycin)对多发性骨髓瘤(multiple myeloma,MM)细胞U266增殖抑制作用。方法以不同终浓度(50、100、200、300、400μg/mL)的褐藻糖胶及不同终浓度(50、100、200、300μg/mL)的褐藻糖胶联合终浓度10nM雷帕霉素作用于U266细胞24h,用CCK-8(cell counting kit-8)法检测细胞增殖抑制作用。结果不同终浓度(50、100、200、300、400μg/mL)的褐藻糖胶作用于U266细胞24h后,其抑制率分别是(9.46±0.04)%、(14.08±0.08)%、(19.97±0.10)%、(25.38±0.13)%、(29.22±0.16)%;不同终浓度(50、100、200、300μg/mL)的褐藻糖胶联合终浓度10nM雷帕霉素作用于U266细胞24h后,其抑制率分别是(15.45±0.41)%、(28.25±0.25)%、(40.88±0.30)%、(56.99±0.18)%。结论不同终浓度的褐藻糖胶均能抑制U266细胞的增殖,并随其浓度增加,抑制率逐渐增大,细胞的增殖抑制作用呈浓度依赖性增强;褐藻糖胶联合雷帕霉素作用于U266细胞比单独用褐藻糖胶抑制率高。 Objective To investigate the inhibitory effect of fucoidan combined with rapamycin on proliferation of U266, multiple myeloma cell. Methods U266 cells were put into solutions of fucoidan of different final concentrations(50μg/mL,100txg/mL ,200μg/mL ,300μg/mL,400μg/mL) and different final fueoidan concentrations ( 50μg/ml, 100μg/ml, 200μg/ml, 300μg/ ml) combined with 10nM rapamyein for 24 hours, and the proliferation inhibition effect was tested by cell counting kit-8 (CCK-8). Re- sults The inhibitory rate of different final fueoidan concentration (50μg/mL, 100μg/mL, 2001xg/mL, 300μg/mL,400μg/mL)on 15266 cells for24 hours were(9.46 ±0.04)% ,(14. 08 ±0.08)% ,(19. 97 ±0. 10)% ,(25.38 ±0. 13)% and(29. 22 ±0. 16)% , respectively. The proliferation inhibition rate of fueoidan of different final eoneentrations(50μg/ml,100μg/ml,200μg/ml,300μg/ml) combined with 10nM rapamyein on U266 cells were ( 15.45 ± 0. 41 ) %, ( 28.25 ± 0. 25 ) %, ( 40. 88 ± 0. 30 ) % and ( 56. 99 ± 0. 18)%. Conclusion The proliferation of U266 cells can be inhibited by fueoidan solution of different final concentration and the inhibitory effect gets stronger with the increasing of the concentration, which indicates that there is a positive concentration dependence in terms of proliferation inhibitory effect. Another conclusion is that fueoidan combined with rapamyein has a stronger inhibitory effect on proliferation of U266 cells than fueoidan applied only.
