摘要
目的探讨靶向下调谷胱甘肽过氧化物酶1(GPX1)对人食管鳞状细胞癌细胞株(EC9706)增殖和顺铂敏感性的影响。方法合成针对GPX1的siRNA,分别用Lipofectamine 2000脂质体瞬时转染人食管鳞状细胞癌细胞株EC9706。Western Blot法检测转染后GPX1蛋白表达,用MTS检测法测定转染后细胞的增殖活力和顺铂敏感性。结果 siRNA干扰EC9706细胞后在36、48、60及72 h对细胞增殖活性较对照组明显抑制(P<0.05)。经siRNA干扰GPX1表达下调,在由不同浓度的顺铂作用后同对照组相比,细胞对顺铂的敏感性明显上升(P<0.05),[IC50(si GPX1)=3.8μmol/L,IC50(阴性对照)=6.1μmol/L,IC50(空白对照)=6.9μmol/L]。结论 siRNA下调GPX1表达可以抑制人食管鳞状细胞癌细胞株EC9706的增殖能力,以及上调其对顺铂的敏感性。
Objective To explore the effect of down - regulated GPX1 gene on the proliferation and the cisplatin sensitivity of esophageal squamous cell carcinoma EC9706. Methods Lipofeetamine 2000 liposomes were used to transfeet human squamous cell car- cinoma cell line EC9706 transiently for GPX1 synthetized siRNA. GPX1 protein expression was measured by Western Blot,proliferation of cell and sensitivity of cisplatin were measured by MTS. Results The proliferation were inhibited than that in control group when EC9706 were interfered with siRNA at 36 h ,48 h,60 h and 72 h ( P 〈 0.05 ). Down - regulated by siRNA interference GPX1 after different concentrations of eisplatin as compared with that in control group, cell sensitivity to eisplatin significantly increased ( P 〈 0.05 ), ( ICS0 ( siGPXI ) = 3.8, ICS0 (Scramble) = 6.1 umol/L, ICS0 ( Blank ) = 6.9 umol/L). Conclusion siRNA down - regulated GPX1 gene can inhibit the proliferation of human esophageal squamous cell carcinoma cell line EC9706 and increase its sensitivity to eisplatin.
