天降血栓通丸早期干预NF-κB信号通路对糖尿病大鼠大血管病变的影响 被引量:1

Effect of early intervention of Tianjiang Xueshuantong pill on NF-κB signal pathway of macroangiopathy in type 2 diabetic rats

摘要 目的:研究天降血栓通丸早期干预的方法能否减少实验大鼠主动脉NF-κB蛋白因子及其下游相关细胞因子的表达,进而延缓糖尿病大血管病变的发生发展。方法:将90只wistar大鼠随机分为空白组(15只),造模组(75只),造模组随机分为模型组,天降血栓通丸早期组、中期组、晚期组与盐酸吡格列酮组,予药物干预,观测血糖、MCP-1、MMP-9,NF-κB。结果:与模型组相比,天降血栓通丸各组血糖无明显差异(P>0.05);天降血栓通丸早期、中期给药组与模型组比较,MCP-1降低,差别具有统计学意义(P<0.05);天降血栓通丸早期给药组与西药组比较,MCP-1表达降低,差别具有统计学意义(P<0.05);与模型组相比,天降血栓通丸早期、中期给药组NF-κB及MMP-9蛋白表达下调明显(P<0.05),其中天降血栓通丸早期组下调最显著。结论:天降血栓通丸早期干预有效降低NF-κB调控的MCP-1及MMP-9蛋白水平表达,可能是其延缓2型糖尿病大鼠大血管病变发生发展的重要机制之一。 Objective: To research whether the early intervention of pill can reduce the expesssions of experi- mental rat's aortic NF - kappa B protein factor and its related downstream cytokines, in order to delay the occur- rence and development of diabetic macroangiopathy. Methods: 90 Wistar rats were divided randomly into control group (15 rats), modeling group (75 rats). The modeling group was divided again randomly into model group, early - , mid - , and late - term intervenient group of pill , pioglitazone hydrochloride group. Given drug for inter- vention, blood glucose, monocyte chemotactic protein - 1 ( MCP - 1 ), matrix metalloproteinases - 9 ( MMP - 9 ), nuclear factor - kappa B ( NF - κB) were observed. Results : Compared with the model group, the blood glucose of early - , mid - , and late - term intervenient group of pill had no significant difference ( P 〉 0.05 ) ; compared medicine group, the MCP - 1 expression decreased, the difference was statistically significant (P 〈 0. 05 ) ; com- pared with the model group, the protein expressions of NF - KB and MMP - 9 of early - , and mid - term interven- ient group of pill were lowered obviously(P 〈 0. 05), and among them, it is the most obvious in early -term in- tervenient group of pill. Conclusion: The early intervention of pill can effectively reduce the expression of MCP - 1 and MMP - 9 protein regulated by NF - kappa B, which may be one of the important mechanisms to retard the occurrence and development of maeroangiopathy in type 2 diabetic rats.
