摘要
目的观察丹参酮Ⅰ(T-Ⅰ)对于放射性膀胱炎(RIBI)的保护作用。方法体内实验:雌性ICR小鼠随机分为对照组1(control)、对照组2(T-Ⅰ)、实验组1(R)、实验组2(R+T-Ⅰ),每组给予丹参酮Ⅰ或橄榄油腹腔注射两周后接受X射线照射,1周或12周后收集小鼠膀胱组织,比较各组膀胱组织内超氧化物歧化酶(SOD)及丙二醛(MDA)水平及组织病理学评分;体外实验:人膀胱永生化上皮细胞(SV-huc-1)分为对照组1(control)、对照组2(T-Ⅰ)、模型组1(R)、模型组2(R+T-Ⅰ),每组利用丹参酮Ⅰ或其溶剂处理24h后接受X射线照射,比较各组细胞活力及细胞内活性氧水平。结果体外实验中,利用T-Ⅰ预处理可以降低X射线导致的膀胱上皮细胞活力下降,并且可以减少细胞内的活性氧水平。在体内实验中,T-Ⅰ治疗能显著减轻RIBI早期膀胱组织损伤,降低组织内氧化应激水平,并改善了RIBI晚期膀胱纤维化程度。结论 T-Ⅰ可通过抗氧化活性在放射性膀胱炎中起到保护作用。
Objective To investigate the protective effect of tanshinone- I (T- I ) on radiation-induced bladder injury (RIBI). Methods In vivo study:The female ICR mice were divided into 4 groups:control group 1 (control), control group 2 (T- I ), model group ] (R), and model group 2 (R+T- I ). Mice in the T- I and R+T- I groups received intraperitoneal in- jection of T- I , while mice in the control group and R group received olive oil. After 2 weeks, the mice were exposed to radia- tion. Urinary bladder samples were collected after 1 week or 1B weeks. Superoxideismutase (SOD), malondialdehyde (MDA) and bladder histopathology score were compared among the groups. In vitro study: the human uretherial cell line (SV-huc-1) was divided into 4 groups:control group 1 (control), control group 2 (T- I ), model group 1 (R) and model group 2 (R--T- I ). Group T- I and R--T- I were treated with T- I , and group R and R+T- I were exposed to radiation after 24 h. Cell vi- ability and reactive oxygen species (ROS) were compared among all groups. Results T- I attenuated radiation-induced via- bility decrease of urothelial cells and ROS generation in vitro. In vivo, treatment of T- I dramatically prevented radiation-in- duced histological damage and reduced oxidative stress in the acute phases of RIBI, while the bladder fibrosis was significantly alleviated in the chronic phase. Conclusion T- I plays a protective role in RIBI via its antioxidant activity.
