丹参注射液对骨性关节炎模型兔膝关节软骨细胞p-IκBα表达影响的实验研究

Experimental Study on Effect of Danshen( Salvia miltiorrhiza)Injection( 丹参注射液) on p-IκBα Protein Expression in Knee Articular Chondrocytes of Rbbits wtih Osteoarthritis

目的:观察丹参注射液对体外培养骨性关节炎模型兔膝关节软骨细胞p-IκBα表达的影响。方法:取12只6月龄新西兰大白兔关节软骨作体外细胞培养,培养后软骨细胞随机分为正常组、模型组、玻璃酸钠组、丹参组4组。采用NO诱导凋亡,各组给予相应处理24 h,Western blot法检测各组软骨细胞p-IκBα的表达。结果:与正常组比较,模型组软骨细胞p-IκBα蛋白明显增强,差异有显著统计学意义(P<0.01);玻璃酸钠组、丹参组p-IκBα蛋白相对表达量明显低于模型组,差异有显著统计学意义(P<0.01);丹参组IκBα蛋白相对表达量明显低于玻璃酸钠组,差异有显著统计学意义(P<0.01)。结论:丹参注射液可以通过抑制IκBα磷酸化,从而抑制NF-κB信号通路的激活,保护软骨下骨及软骨细胞,有效防治OA。

Objective： To observe the effect of Salvia miltiorrhiza Injection on p - I kappa B expression in rabbits＇ knee articular chondrocytes cultured in vitro. Methods： Taking 12 New Zealand white rabbits＇ knee ar- ticular cartilage cells cultured in vitro, chondrocytes cultured in three generations, were randomly divided into normal group, model group, sodium hyaluronate group, Salvia mihiorrhiza group four groups. Using SNP to induce an external model of osteoarthritis, articular chondrocytes of each group were cultured with the correspond- ing medicine for 24 h, and the Western blot method was used to determine the expression of the chondrocytes p -IKBot. Results：Compared with normal group, the p - I kappa B expression in chondrocytes of model group were significantly enhanced and the difference was statistically significant （P 〈 0.01 ）, the expression in sodium hyalate group and Salvia mihiorrhiza group was slighter than that in model group , the differences were both statis- tically significant（ P 〈 0. 01 ）, the p - I kappa B expression in chondrocytes of Salvia mihiorrhiza group was lower to compare with sodium hyalate group, the difference was statistically significant （P 〈 0. 01 ）. Conclusion： Sal- via mihiorrhiza Injection can inhibit the activation of NF - kappa B signaling pathway by inhibiting the phospho- rylation of I kappa B alpha, and protect the subchondral bone and cartilage cells, so as to prevent and treat OA effectively.
