摘要
目的 通过体外无菌培养Sgc-7901细胞,观察加入冬凌草甲素后,Sgc-7901细胞炎性因子表达的影响,评价冬凌草甲素对Sgc-7901细胞炎性因子的变化差异,探讨冬凌草甲素抗肿瘤的机制,为临床治疗胃癌提供新的理论依据.方法 无菌培养Sgc-7901细胞,CCK-8实验筛选冬凌草甲素作用24h后抑制Sgc-7901细胞增殖的最佳作用浓度;液相芯片分析系统测定冬凌草甲素对Sgc-7901细胞IL-2、IL-4、IL-6、IL-10以及TNF-α炎性因子的影响.结果 CCK-8实验结果显示,冬凌草甲素能抑制Sgc-7901细胞增殖,并且呈剂量依赖性,且IC50分别是66.88μm;液相芯片分析系统检测冬凌草甲素处理前后Sgc-7901细胞IL-2、IL-4、IL-6、IL-10以及TNF-α炎性因子差异表达结果显示:冬凌草甲素组与空白对照组比较,部分炎性因子表达下调.结论 冬凌草甲素可以抑制Sgc-7901细胞的增殖,下调了Sgc-7901细胞中IL-6等炎性因子的分泌,从而达到抗肿瘤的作用.
Objective To observe the effect of the expression of inflammatory cytokines in Sgc-7901 after adding the oridonin and to evaluate oridonin on the changes of inflammatory cytokines in Sgc-7901 and the effects of oridonin on anti-tumor mechanism through SGC-7901 cells cultured in vitro by sterile,then provide a new theoretical basis for the clinical treatment of gastric cancer. Methods Sgc-7901 cells fostered in sterile culture, through CCK-8 experiment,the optimal concentration to inhibit the proliferation of Sgc-7901 cells were screened to oridonin after 24h. Liquid chip analysis system for determination of oridonin on the effect of IL-2,IL-4,IL-6,IL-10 and TNF-α in Sgc-7901 cells. Results CCK-8 experiment results showed that oridonin could inhibit the proliferation of Sgc-7901 cells in a dose-dependent manner,and the IC50 was 66.88μm. The results of Liquid chip analysis system that to test the differential expression of IL-2,IL-4,IL-6,IL-10 and TNF-α after oridonin treated Sgc-7901 cells before and after showed that partial inflammatory factors expression down regulated,the oridonin group with the blank control group were compared. Conclusion Oridonin can inhibit the proliferation of Sgc-7901 cells,and reduce the secretion of IL-6 and other inflammatory factors in Sgc-7901 cells,so as to achieve anti-tumor effect.
