Optimization of phytosterol extraction from physalis fruit by response surface method

摘要 对酸浆果实中总甾醇含量采用超声波法提取,以β-谷甾醇为标准品配制标准溶液,绘制标准曲线,标准曲线的线性回归系数为0.9965。以提取到的甾醇含量(mg)为评价指标,对酸浆果实中甾醇的提取工艺,如提取温度、料液比、乙醇体积分数和提取时间进行单因素和响应面优化试验,确定最佳提取工艺为:提取温度58℃、提取时间30min、提取剂浓度为100%乙醇溶液、料液比为1∶44(g∶m L)。对响应面优化的实验结果进行验证试验,按照响应面试验优化得到的提取条件:提取温度58℃、料液比1∶44(g∶m L)、乙醇体积分数100%、提取时间30min,得到结果为1.774‰,与响应面预测结果 1.786‰相差较小,故响应面得到的结果真实可靠。 In this paper, total phytosterols were extracted from physalis fruit by ultrasonic wave method. Standard solution was prepared using beta sitosterol as the standard material, and the standard curve was drawn with a regression coefficient of 0.9965. The extracted sterol content (rag)was used as the evaluation index, the extraction process of sterols from physalis fruit were optimized by response surface method, and the extraction process such as extraction temperature, solid-liquid ratio, ethanol concentration and extraction time were selected as the single factors to complete the single factor test. As a result, the optimal technological conditions were as follow : extraction temperature was 58℃, extraction time length was 30 min, extract concentration was 100% ethanol solution, solid- liquid ratio was 1 : 44 (g : mL). The experimental results were verified by experiment, and the result was compared with the predicted result, the extraction yield from experiment was 1.774% which has a small difference with the prediction result (1.786%)from the response surface method. In summary, the result obtained from the response surface method is real and reliable
