Smad4、TGF-β1、TGF-βR1在乳腺癌细胞中的表达和意义 被引量:4

Expressions and significance of Smad4,TGF-β1,and TGF-βR1 in breast cancer cells

摘要 目的探究Smad4、转化生长因子-β1(TGF-β1)、转化生长因子-β受体1((TGF-βR1)在乳腺癌细胞中的表达和意义。方法选取2014年1月-2016年1月在该院确诊为乳腺浸润性导管癌并行手术治疗的女性患者60例,收集其标本作为观察组。另取正常乳腺组织20例作为对照组。采用免疫组化技术检测组织中的Smad4、TGF-β1和TGF-βR1,比较乳腺浸润性导管癌和正常乳腺组织中上述指标的表达差异及影响表达的因素。结果观察组TGF-β1和TGF-βR1的阳性表达率高于对照组,Smad4阳性表达率低于对照组,差异均有统计学意义(均P<0.05)。Smad4、TGF-β1和TGF-βR1在乳腺癌中的表达与年龄无关,而同分化程度和TNM分期有关,分化程度低、TNM分期高的标本TGF-β1和TGF-βR1的阳性表达程度高于分化程度高、TNM分期低的标本;分化程度低、TNM分期高的标本Smad4的阳性表达程度低于分化程度高、TNM分期低的标本,差异均有统计学意义(均P<0.05)。TGF-β1和TGF-βR1在乳腺癌组织中的表达呈正相关(r=0.647,P<0.05),而Smad4与TGF-β1、TGF-βR1的表达均呈负相关(r=-0.329和-0.426,均P<0.05)。结论 TGF-β1和TGF-βR1在乳腺浸润性导管癌组织中表达显著增加,而Smad4表达下降,表达程度均与分化程度和TNM分期有关。 Objective To explore the expressions and significance of Smad4,transforming growth factor-β1( TGF-β1),and transforming growth factor-β receptor 1( TGF-βR1) in breast cancer cells. Methods A total of 60 patients diagnosed as breast invasive ductal carcinoma definitely in the hospital from January 2014 to January 2016 were selected as observation group. A total of 20 cases with normal breast tissue were selected as control group. The expressions of Smad4,TGF-β1,and TGF-βR1 were tested through immunohistochemical technique,the expression differences and influencing factors of their expressions were compared between the two groups. Results The positive expression rates of TGF-β1 and TGF-βR1 in observation group were higher than those in control group,and the positive expression rate of Smad4 in observation group was lower than that in control group,there were statistically significant differences( P〈0. 05). The expressions of Smad4,TGF-β1,and TGF-βR1 in breast cancer had no relation with age,while they had relations with degrees of differentiation and TNM staging. The positive expression rates of TGF-β1 and TGF-βR1 in patients with low degrees of differentiation and high TNM staging were higher than those with high degrees of differentiation and low TNM staging,while the positive expression rate of Smad4 in patients with low degrees of differentiation and high TNM staging was lower than that with high degrees of differentiation and low TNM staging,there were statistically significant differences( P〈0. 05). The expression of TGF-β1 in breast cancer was positively correlated with the expression of TGF-βR1( r = 0. 647,P〈0. 05). The expression of Smad4 in breast cancer was negatively correlated with the expressions of TGF-β1and TGF-βR1( r =-0. 329,-0. 426,P〈0. 05). Conclusion The expressions of TGF-β1 and TGF-βR1 significantly increase in the tissue of breast invasive ductal carcinoma,while the expression of Smad4 decreases,which are related to the degree of differentiation and TNM staging.
