水通道蛋白-1在藏族宫颈鳞癌患者中的表达及病理学意义 被引量:1

Expression and pathological significance of aquaporin-1 in cervical squamous cell carcinoma of Tibetan women

摘要 目的探讨水通道蛋白-1(AQP1)在青海省藏族宫颈鳞癌患者中的表达及病理学意义。方法收集2014年11月-2015年6月在青海大学附属医院和青海省人民医院住院行手术治疗的藏族宫颈鳞癌(Ιa1期~Ⅱa期)患者50例,采用RT-q PCR方法检测宫颈鳞癌组织和癌旁组织中AQP1的基因表达水平,采用免疫组化法检测分析其形态学表达定位和意义。结果RT-q PCR方法检测结果显示,藏族宫颈鳞癌患者患癌组织AQP1 mRNA的表达量高于癌旁组织,分别为5.72±1.05和4.05±0.83,差异有统计学意义(t=7.89,P<0.01)。免疫组化法检测结果与RT-q PCR法检测结果一致,AQP1在宫颈鳞癌组织中的表达量高于癌旁组织,差异有统计学意义(t=2.02,P<0.05)。免疫组化结果显示,在宫颈鳞癌肿瘤细胞的细胞膜及细胞质中有AQP1的表达,表现为明显棕黄色,同时肿瘤间质血管内皮细胞中也有显著表达。结论 RT-q PCR和免疫组化法均证实AQP1在藏族宫颈鳞癌患者中的表达上调,在肿瘤细胞的细胞膜及细胞质中高表达,在肿瘤间质血管内皮细胞中亦显著表达。 Objective To explore the expression and pathological significance of aquaporin-1( AQP1) in cervical squamous cell carcinoma of Tibetan women in Qinghai province. Methods A total of 50 cases with cervical squamous cell carcinoma( stage Ιa1 ~ Ⅱa) treated in Affiliated Hospital of Qinghai University and Qinghai Provincial People's Hospital from November 2014 to June 2015 were selected,gene expression levels of AQP1 in cervical squamous cell carcinoma tissue and adjacent normal tissue were detected by RT-q PCR,immunohistochemical method was used to detect the morphological expression localization and significance. Results The results of RT-q PCR showed that the expression level of AQP1 mRNA in cervical squamous cell carcinoma tissues and the adjacent normal tissues were( 5. 72±1. 05) and( 4. 05±0. 83),respectively,there was statistically significant difference( t = 7. 89,P〈0. 01). The test results of immunohistochemical method were consistent with RT-q PCR test results,the expression level of AQP1 in cervical squamous cell carcinoma tissue was statistically significantly higher than that in adjacent normal tissue( t = 2. 02,P〈0. 05). The results of immunohistochemical method showed that there were AQP-1 expressions in cell membrane and cytoplasm of cervical squamous cell carcinoma cells,which was significantly brown-yellow,and the expression of AQP1 was also significant in stromal vascular endothelial cells. Conclusion RT-q PCR and immunohistochemical method show the up-regulation of AQP1 in cervical squamous cell carcinoma tissue,AQP1 highly expresses in cell membrane and cytoplasm of cervical squamous cell carcinoma cells and stromal vascular endothelial cells.
