Notch1,Jagged1在结肠癌中的表达及意义

The Expression and Significance of Notch1 and Jagged1 in Colon Cancer

摘要 目的探讨Notch1,Jagged1在结肠癌组织中的表达及其相关性,分析其临床病理学意义。方法随机选取51例经手术切除的结肠癌组织石蜡组织标本及51例癌旁结肠组织石蜡标本,所有被切除的组织均被病理证实,患者术前均无放疗和化疗等治疗。以癌旁组织作为对照组,通过免疫组化SP法检测Notch1,Jagged1在结肠癌及癌旁组织中的表达及其定位,并探究两种因子之间的表达相关性。结果 Notch1和Jagged1均主要表达于细胞膜及细胞质中,且在结肠癌组织中的阳性表达率均显著高于癌旁组织。两者的表达与结肠癌的分化程度、临床分期、淋巴结转移密切相关(P<0.05),Notch1和Jagged1在结肠癌组织中的表达均呈正相关关系(r=0.499,P<0.05)。结论 Notch1及Jagged1在结肠癌组织中的高表达与结肠癌的发生发展及转移密切相关,检测两者的表达对结肠癌的诊断、治疗有重要的临床意义。 Objective To investigate the expression of Notch1,Jagged1 in colon cancer tissues and their correlation.Methods All the samples were randomly selected from the pathology,including 51 cases of surgically resected cancer tissue paraffin tissue samples and 51 cases of paracancerous tissue paraffin specimens,and all the tissue cells were pathologically confirmed.All the specimens had not received any chemotherapy or radioactive treatment before surgery.The adjacent tissues were used as control group.The expression and localization of Notch1 and Jagged1 in colon cancer and adjacent tissues were detected by immunohistochemical SP method.Results Notch1 and Jagged1 were mainly expressed in the cytoplasm and cell membrane,and the rate of positive expression in colon cancer was significantly higher than that in adjacent tissue.The expression of the two factors was closely related with the differentiation of colon cancer,clinical stage and lymph node metastasis(P〈0.05).There was significantly positive correlation between Nocth1 and Jaggedd1 in human colon cancer(r=0.499,P〈0.05).Conclusion The overexpression of Notch1 and Jagged1 are closely correlated with carcinogensis,progression and metastasis of colon cancer,which promote the occurrence and the development of colon cancer.Detecting the expression of these two factors has important clinical significance in the diagnosis and treatment of colon cancer.
