摘要
目的:探讨平肝止痛颗粒治疗偏头痛的作用机制,研究平肝止痛颗粒对肝阳上亢型偏头痛大鼠模型血浆中降钙素基因相关肽(Calcitonin Gene-related Peptide,CGRP)和五-羟色胺(Five-serotonin,5-HT)含量的影响。方法:采用皮下注射硝酸甘油(Nitroglycerin,NTG)注射剂复制实验性偏头痛大鼠模型,灌胃平肝止痛颗粒治疗后,采用酶联免疫法测定血浆CGRP和5-HT含量。结果:皮下注射硝酸甘油(NTG)注射剂偏头痛大鼠模型血浆中CGRP含量与空白组比较有明显升高,而5-HT含量与空白组比较有不同程度的升高。平肝止痛颗粒24 g生药/kg,12 g生药/kg,6 g生药/kg剂量均可明显降低偏头痛模型大鼠血浆中CGRP、5-HT含量水平。讨论:平肝止痛颗粒可能通过对血管活性物质CGRP与5-HT水平的调节,来改善血管的异常舒缩状况,抑制神经性炎性反应,从而达到缓解疼痛作用和对偏头痛的治疗作用。
Objective: To discuss the mechanism of Pingganzhitong granules in the treatment of migraine and study its effect on the content of calcitonin gene-related peptide (CGRP) and 5-hydroxytryptamine (5-HT) in the plasma of rats with hyperactivity of liv- er-yang and migraine. Methods :The subcutaneous injection of nitroglycerin was performed to duplicate the rate model of experi- mental migraine. After the irrigation of Pingganzhitong granules, the enzyme-linked immunosorbent assay was employed to detect the content of CGRP and 5-HT in the plasma. Results : After the subcutaneous injection of nitroglycerin (NTG) in rat model of mi- graine, the content of CGRP in the plasma was significantly increased compared with the control group, while the content of 5-HT was increased to a certain higher extent compared with the control group. The dosage of Pingganzhitong granules with 24 g crude drugs/kg, 12 g crude drugs/kg and 6 g crude drugs/kg could significantly reduce the content of CGRP and 5-HT in the plasma of rat model of migraine. Conclusion:By regulating the content of vasoactive substances of CGRP and 5-HT, the Pingganzhitong granules can improve the abnormal vasodilation, inhibit the neurogenic inflammation and thus relieve the pain and achieve the treatment of migraine.
