加味当归补血汤对化疗小鼠红白细胞免疫功能的影响

Effect of Jiawei Danggui Buxue decoction on red and white cell immune function in mice after chemotherapy

目的观察加味当归补血汤对化疗后荷瘤小鼠免疫功能的调节作用,筛选出治疗效果最佳剂量。方法以H22荷瘤小鼠左下腹腔注射环磷酰胺为动物模型,将60只昆明种小鼠随机分为6组,即空白对照组、荷瘤组、模型组、加味当归补血汤高、中、低剂量组。连续用药14 d,模型组和加味当归补血汤组在给药第12天开始给环磷酰胺(CY)3 d,第15天解剖小鼠,检测各项试验指标。结果与对照组比较,加味当归补血汤能够提高外周血中RBC、WBC、Hb水平;能提高经CY化疗后EPO的浓度,提高造血祖细胞的增值能力;能够改善经化疗后小鼠血清中T淋巴细胞亚群失调状态以及T淋巴细胞增殖程度,从而提高化疗后小鼠的免疫功能。差异有统计学意义(P<0.05),中剂量效果最佳(P<0.01)。结论加味当归补血汤各剂量能有效的抑制CY化疗所致的骨髓抑制和免疫抑制的毒副作用,从而提高其免疫功能,中剂量效果最佳。

Objective To observe the regulatory effect of Jiawei Danggui Buxue decoction on the immune function of tumor bearing mice after chemotherapy, and screen out the best treatment dose. Methods The H22 tumor bearing mice were injected with cyclophosphamide into the left lower abdominal cavity as animal model, 60 Kunming mice were randomly divided into 6 groups： blank control group, tumor group, model group, Jiawei Danggui Buxue decoction high, middle and low dose group. Continuous administration for 14 d, the model group and the Jiawei Danggui decoction group were treated with cyclophosphamide（CY） 3d after the twelfth day, and the mice were dissected in the fifteenth days, test the experiment index. Results Compared with the control group, Jiawei Danggui Buxue decoction can enhance the level of Hb, RBC, WBC in the peripheral blood, improve the concentration by CY EPO after chemotherapy, improve hematopoietic progenitor cells proliferation ability, be improved after chemotherapy in mice serum T lymphocyte subsets imbalance and proliferation of T lymphocytes, thereby enhancing immune chemotherapy the function of mice. The difference was statistically significant（P〈0.05）, and the dose effect was the best（P〈0.01）. Conclusion Jiawei Danggui Buxue decoction each dose can inhibit bone marrow and immune suppression induced by CY chemotherapy effective suppression of side effects, so as to improve the immune function, dose effect is the best.
