高胰岛素和高血糖对糖尿病患者IGF2BP2基因表达的影响

摘要 目的研究胰岛素量与血糖量对1型和2型糖尿病的患者IGF2BP2基因表达的影响。方法自白细胞提取总mRNA,作为模板进行反转录得到cDNA,以Real-Time PCR方法测定目的基因的表达量。结果测定了18例Ⅰ型糖尿病患者、25例2型糖尿病患者和20名正常人中的IGF2BP2基因表达量。结论该研究在Ⅰ型和2型糖尿病的患者、正常人群中测定了IGF2BP2基因的表达量水平,发现胰岛素值低、血糖高的1型糖尿病组的IGF2BP2基因表达正常,胰岛素与正常组无差异、血糖高的2型糖尿病组IGF2BP2基因高表达,推测胰岛素值、血糖值与IGF2BP2基因无直接联系,但是IGF2BP2可能与胰岛素抵抗有关。 Objective To research the effect of insulin dose and blood glucose dose on the IGF2BP2 gene expression of type 1 diabetes patients and type 2 diabetes patients. Methods The total mRNA was extracted by white cell as templets for reverse transcription thus obtaining cDNA, and the expression level of target gene was measured by the Real-Time PCR method. Results The IGF2BP2 gene expression levels of 18 cases of patients with type 1 diabetes, 25 cases of patients with type 2 diabetes and 20 cases of normal people were measured. Conclusion The measurement shows that the IGF2BP2 gene expression in the type 1 diabetes group with low insulin value and high blood glucose is normal, and there is no difference in the insulin compared with the normal group thus speculating that the insulin value, blood glucose value have no direct correlation with the IGF2BP2 gene, but the IGF2BP2 mac be related to the insulin resistance.
