摘要 目的:观察BAG3蛋白在胶质瘤中的表达,及其ser187位点磷酸化与胶质瘤间的相关性。方法:收集胶质瘤组织标本,进行分子生物学实验,观察BAG3蛋白在胶质瘤组织中的表达情况及BAG3蛋白ser187位点磷酸化对细胞侵袭的影响。结果:(1)IHC:BAG3在Ⅳ级胶质瘤组织表达显著高于正常脑组织及Ⅰ、Ⅱ级胶质瘤组织(P<0.05)。(2)Western Blot:BAG3蛋白在Ⅱ、Ⅲ、Ⅳ级别胶质瘤表达量,显著高于正常脑组织(P<0.05),且相对于Ⅱ级胶质瘤,在Ⅲ、Ⅳ级胶质瘤中表达显著增高(P<0.05),而在Ⅲ级、Ⅳ级胶质瘤组织中表达量之间无显著性差异(P>0.05)。(3)三维Matrigel transwell转移小室实验,S187A-BAG3蛋白过表达U87细胞侵袭Matrigel模拟基质膜的细胞数显著高于U87细胞和S187D-BAG3过表达U87细胞。结论:BAG3蛋白胶质瘤组织中表达量高,且在Ⅳ级胶质瘤组织中,BAG3蛋白的表达明显高于Ⅰ、Ⅱ级及正常脑组织,且S187位点磷酸化能抑制U87细胞的侵袭。 Objective: to observe the BAG3 protein expression in glioma,and its ser187 phosphorylation associated with glioma.Methods:Collected 94 glioma tissue and 56 normal brain tissue,use molecular biological technique,observe the expression of BAG3 protein in the gliomas tissue and effects of the BAG3 protein ser187 phosphorylation on cell invasion.Results:① The expression level of BAG3 in IV glioma group is significantly higher than normal brain tissue and Ⅰ,Ⅱ grade glioma tissues(P〈0.05).②Western Blot results: the expression level of BAG3 protein in Ⅱ,Ⅲ,Ⅳ glioma ,which was significantly higher than that of normal brain tissue (P〈0.05),the level of expression in and grade Ⅲ or IV glioma ,which was significantly higher than that of grade Ⅱ (P〈0.05),expressed in level Ⅲ and Ⅳgrade has no significant differences (P〉0.05).③The 3-d Matrigel transwell transfer chamber experiment results,the results hint S187A-BAG3 protein overexpression of U87 cell invasion on Matrigel simulation matrix membrane of cells was significantly higher than that of U87 cells and S187D-BAG3 express U87 cells.Conclusion:The level of BAG3 protein expression in IV glioma tissues,which is significantly higher than I,II and normal brain tissue,and S187 phosphorylation sites can inhibit U87 cells invasion.
