摘要
建立了HPLC内标法测定苦荞中黄酮类物质的含量。选用芦丁作为检测目标。色谱条件:日本岛津C18柱(250mm×4.6mm i.d.,5mm),柱温:30℃,流动相:乙腈-0.1%磷酸溶液梯度洗脱,流速1.0m L·min^(-1),检测波长:254nm,在0.005~0.2mg/ml线性关系良好。平均加标回收率为100.3%(RSD=4.60%)。结果表明,该方法准确、精密、简便、快速。可作为苦荞中测定总黄酮含量的方法。
The content of flavonoids in tartary buckwheat was determined by HPLC internal standard method. Use Rutin as the target, chromatographic conditions: The Shimadzu-C18 pillar (250mm×4.6mm i.d.,5um), column temperature: 30℃, Flow phase: acetonitrile-0.1% phosphate solution gradient elution, Velocity 1.OmL ·min-1, detecting wavelength: 254nm, in 0.005~0.2mg/ml linear relationship is good. The average recovery rate is 100.3% (RSD= 4.6%). The results show that the method is simple, precise and fast. It can be used as a method for determination of total flavonoids in tartary buckwheat.
