毛梾腋芽诱导关键因子研究与培养体系优化 被引量:1

摘要 [目的]建立毛梾茎段外植体高效腋芽诱导体系。[方法]以毛梾幼嫩茎段作为外植体,对灭菌剂及灭菌时间、采集时期、基本培养基、植物生长调节剂、pH值等腋芽诱导的影响因素进行研究。[结果](1)采用0.1% HgCl_2灭菌4~5min是最佳灭菌剂和灭菌时间;(2)4~5月是外植体较为理想的采集时期;1~3月是最佳采集时期,采用当年生枝条水培后的幼嫩茎段;(3)MS培养基是茎段腋芽诱导的最佳基本培养基;(4)MS培养基添加0.5~1.0 mg·L^(-1)BA、0.1mg·L^(-1) NAA与0.1mg·L^(-1) IBA时,腋芽诱导率最高,分别达85.5%和84.4%;(5)最佳pH值为5.8;(6)光照培养环境更有利于毛梾腋芽诱导及生长。[结论]水培后的幼嫩茎段外植体经0.1%HgCl2灭菌4~5min后,培养在MS添加0.5~1.0mg·L^(-1) BA、0.1mg·L^(-1) NAA与0.1mg·L^(-1) IBA,是毛梾腋芽诱导的最佳培养体系。 [Objective]To establish an efficient system for axillary bud induction using young stem segments as explants of Cornus walteri.[Methods]The paper studied the influence factors which included the disinfectant types and the disinfection time,the collection period of explants,the types of basic culture medium,the combination of plant growth regulators,pH et al.[Result](1)The best disinfectant and disinfecting time was 0.1% mercuric chloride for 4-5min;(2)The better collection period for explants was from April to May;The best collection period was from January to March,using young stem segments as explants after water culture;(3)The optimum medium was MS medium for axillary bud induction;(4)The frequency of axillary bud induction were the highest when the medium supplemented with 0.5-1.0mg·L^-1 BA and 0.1mg·L^-1 NAA and 0.1mg·L^-1 IBA with 85.5% and 84.4%;(5)The optimum pH was 5.8 for axillary bud induction;(6)Light culture was more beneficial for the induction and development of axillary bud.[Conclusion]Using young stem segments as explants after water culture,0.1% mercuric chloride as disinfectant for 4-5min,cultured on MS supplemented with 0.5-1.0 mg·L^-1 BA and 0.1 mg·L^-1 NAA and 0.1 mg·L^-1 IBA,were the best culture system for axillary bud induction of Cornus walteri.
