辣椒品种镇研22种子纯度的SSR分子标记鉴定 被引量:5

摘要 分子标记技术因其具有高效、稳定、可靠等优点越来越多地被应用到种子纯度鉴定中;而基于分子标记的辣椒(Capsicum annuum L.)杂交种子纯度快速检测技术体系的建立对辣椒杂交品种的选育和生产则具有重要的意义。以辣椒杂交品系镇研22及其两亲本为试验材料,利用SSR分子标记技术对辣椒杂交品种间的多态性进行了引物筛选和种子纯度鉴定研究。结果表明,从20对SSR引物中筛选出CAMS-327引物,在亲本和杂交种中表现出稳定的共显性,并对镇研22杂交种进行纯度鉴定,结果为98%,与田间鉴定结果基本一致;SSR标记分析可实现对辣椒杂交品种种子纯度的快速和准确鉴定。 The molecular marker technique has been applied to the identification of seed purity increasingly due to its advantages of being efficient, stable and reliable. DNA-based assessment of seed purity is one of the most important quality control components in hot pepper(Capsicum annuum L.) hybrid seed production. In this study, the seed genetic purity of hot pepper hybrid cultivar Zhenyan 22 was analyzed with SSR marker, genomic DNA from the Zhenyan 22 and its parental lines was screened with 20 pairs of SSR primers, from which one pair of primers CAMS-327 that could perform stable co-dominance in the parental lines and hybrids and produce both female and male parent specific markers was selected for the genetic purity test, and a total of 45 hybrid individuals were analyzed with the identified primer. The results showed that the overall genetic purity of the hybrid seed Zhenyan 22 was at 98% which was relatively consistent with that of the field test. The combination of DNA extraction and SSR marker analysis is a rapid and accurate method for the detection of hybrid hot pepper variety seed purity.
