摘要
目的:对防风二氯甲烷及乙酸乙酯部位化学成分进行研究。方法:采用镜下观察细胞病变效应(CPE)法,以Vero-HSV-1(F),RD-EV71,Vero-RSV体外感染模型,对防风不同极性部位进行体外抗病毒活性初步筛选,发现防风二氯甲烷及乙酸乙酯部位具有体外抗HSV-1的活性,采用硅胶柱色谱、凝胶柱色谱、液相制备、重结晶等手段对其进行分离纯化,通过理化性质,MS,1H-NMR和13C-NMR等谱学技术进行结构鉴定。结果:从中分离得到11个化合物,分别鉴定为:单棕榈酸甘油酯(1),2-丁氧基乙基亚油酸(2),3'(R)-(+)-亥茅酚(3),(-)-marmesin(4),(6Z,9Z)-十七烷(5),32,33,34-三甲基-16-烯-3-O-β-D-吡喃葡萄糖苷(6),亥茅酚苷(7),香草酸(8),升麻素(9),5,7-二羟基-2-甲基色原酮(10),伞形花内酯(11)。结论:化合物2,5,6,10为首次从该属植物中分离得到。
Objective:To study the chemical constituents of the methylene chloride and ethyl acetate extract of Saposhnikovia Radix.Method:The activity of anti-HSV-1 was screened by CPE method with Vero-HSV-1(F),RD-EV71 and Vero-RSV in vitro infection models and the compounds were isolated and purified by silica gel column chromatography,gel column chromatography,HPLC preparation and recrystallization.The structures were elucidated by the physical and chemical properties,MS,1H-NMR and13C-NMR spectroscopy.Result:The11 compounds were isolated and their structures were identified asglyceroylmonopalmitate(1),2-butoxyethyl linoleate(2),3’(R)-(+)-hamaudol(3),(-)-marmesin(4),(6Z,9Z)-heptadecadiene(5),32,33,34-trimethyl-bacteriohopan-16-ene 3-O-β-D-glucopyranoside(6),sec-O-glucosylhamaudol(7),vanillic acid(8),cimifugin(9),noreugenin(10) andumbelliferone(11).Conclusion:Compounds 2,5,6 and 10 were isolated from the genus for the first time.
