THE EFFECT OF MYOCARDIAL PROTECTION BY VERAPAMIL AND/OR COENZYME Q_(10) ON ISOLATED RAT HEART

<正> 近年来,心肌保护的研究成为心脏外科的重要课题之一。本实验旨在评价维拉帕米(异搏定)和(或)辅酶Q_(10)加入冷钾停跳液中对心肌的保护效果,为临床应用提供实验依据。

The effect of verapamil and/or coenzyme Q_(10) added to cardioplegic solution to enhance myocardial protection was studied on isolated perfused rat heart model. Thirty-two male Sprague-Dawley rats (280±16g) were randomly divided into four groups (eight in each). Croup Ⅰ, as control, re ceived cold potassium cardioplegic solution, Croup. Ⅱ received the same solution as group Ⅰ plus vera pamil (0.5mg/L), Croup Ⅲ the same solution as group Ⅰ plus coenzyme Q_(10) (60mg/L) and Croup Ⅳ the same solution as group Ⅰ plus verapamil (0.5mg/L) and coenzyme Q_(10) (60mg/L). The recovery percent of myocardial contractility, lactic acid dehydrogenase (LDH) and creatine phoapho kinase (CPK) released, and coronary flow were measured before ischemia and 10 min, 20 min, and 30 min of reperfusion. Ultrastructure of myo cardial tissue was examined before ischemia and 30 min of reperfusion. Results showed that either verapamil or coenzyme Q_(10) added to the cardio plegic solution (group Ⅱ or Ⅲ) had better myocar dial protection than group Ⅰ. With both verapamil and coenzyme Q_(10) added (group Ⅳ), much better protection was obtained.
