摘要
目的:建立内标法测定人血浆中鞣花酸的浓度。方法:选用水杨酸为内标,色谱柱为伊力特SinoChrom ODS柱(4.6mm×150mm,5μm),流动相:乙腈-0.2%磷酸溶液(25∶75);检测波长238nm;流速1ml/min。结果:该方法的线性范围为0.5~10μg/ml,R2=0.9998,鞣花酸保留时间3.9min,平均回收率为99.94±1.22%(n=15),精密度实验RSD≤3.5%。鞣花酸的最低定量浓度为0.5μg/ml,进样量在10~200ng范围内线性关系良好。结论:该方法灵敏较高、专属性较强,可用于临床血药浓度监测和人体药代动力学研究。
Objective:To establish a HPLC method for the determination of Ellagic acid in human plasma.Methods:Salicylic acid was used as the internal standard.The chromatographic column was Elet SinoChrom ODS(4.6mm×150mm,5μm)with a mobile phase of acetonitrile-0.2% phosphoric acid solution(25∶75);the detection wavelength was 238nm;the flow rate is 1ml/min.Results:The linear range of this method was0.5~10μg/ml R2=0.9998.The retaining time of Ellagic acid was 3.9min;the average recovery was 99.94±1.22%(n=15);precision experiment RSD≤3.5%.The detection limit of Ellagic acid was 0.5μg/ml,and the linear relation was good when the sample size was in the range of 10~200ng.Conclusion:The method has high sensitivity and strong specificity,and it can be used for clinical monitoring of Ellagic acid and its pharmacokinetic studies.
