乳杆菌ALAC-3产抑酵母菌活性物质培养基的优化 被引量:1

摘要 从内蒙古传统乳酸发酵食品中筛选出具有较强抑酵母菌作用的一株乳杆菌ALAC-3。从8组适合乳杆菌菌株生长的培养基中筛选出代谢产生抑酵母活性物质的最优培养基,再通过单因素和正交实验,确定培养基中最适宜组分和适宜添加量。结果表明,碳源、氮源对菌株产抑菌物质有显著的影响,最优培养基为:蔗糖25 g/L,蛋白胨∶牛肉膏∶酵母膏(2∶1∶1)20 g/L,K+:Mg2+:Mn2+(10∶3∶1)0.1 mol/L,生长因子乙醇2 m L/L。以此培养基培养ALAC-3,其抑菌圈可达32.68 mm,比培养基优化前(抑菌圈29.91 mm)提高了9.3%。 The inhibiting activity of antiyeast substances produced by Lactobacillus ALAC-3, one strain isolated from traditional dairy fermentation food in Inner Mongolia was studied. In this study, one best was singled out of eight culture media that suited ALAC-3 to grow and produce antiyeast active substances. Then the best components and its volume range were confirmed by Single-Factor experiments and Orthogonal experimental design. Experimental results indicated that the carbon source, nitrogen source significantly affected the inhibiting activity on yeast. When the parameters were composed as follows: 25 g/L of sucrose, 20 g/L of peptone:beef extract: yeast extract (2: 1: 1), 0.1 mol/L of K+-Mg2+-Mn2+(10:3:1 ), 2 mL/L of ethanol as growth factor, the inhibiting activity was maximum with verified value of 32.68 mm, increased by 9.3 % from the initial value of 29.91 mm.
