摘要 等温扩增技术(IAT)作为一种新型的体外核酸扩增技术,与PCR相比,该技术具有特异性强、敏感性高、简单便捷和成本低等优点。目前,等温扩增技术在病原检测方面得以应用,论文对环介导等温扩增(LAMP)、依赖解旋酶的等温扩增(HDA)、依赖核酸序列的等温扩增(NASBA)、链置换等温扩增(SDA)、交叉引物扩增(CPA)5种扩增技术的原理、特点及应用分别予以介绍,为该技术在病原检测的实际应用提供参考。 Isothermal amplification technology is a new type of nucleic acid amplification in vitro that grows rapidly in recent years.Compared with traditional PCR,this technology takes advantages such as low cost, easy-to-operate, higher sensitivity and specificity.Many isothermal amplification technologies are applied to the detection of bacteria, viruses and other pathogens.Here the principle, feature and application of isothermal amplification technologies are reviewed including loop-mediated isothermal amplification (LAMP),helicase-dependent isothermal amplification (HDA), nucleic acid sequence-based amplification (NASBA), strand displacement amplification (SDA), and cross priming amplification (CPA). It provides a reference for the practical applications in the detection of pathogens.
