摘要
目的研究别嘌醇(allopurinol,Allo)联合5-氟尿嘧啶(5-Fu)对肝癌HepG2细胞增殖和凋亡的影响。方法用不同质量浓度和配伍的别嘌醇、5-Fu处理HepG2细胞,用CCK-8法检测细胞增殖,用流式细胞术检测细胞凋亡。结果单用别嘌醇对HepG2细胞增殖有微弱的抑制作用;别嘌醇与5-Fu联用能抑制HepG2细胞的增殖,并具有质量浓度(1~3μg/L)和时间(24,48,72 h)依赖性,两药联用比单用5-Fu时抑制率更高;别嘌醇与5-Fu联用时HepG2细胞的凋亡率为39.17%,高于单用5-Fu的30.86%(P<0.05)。结论别嘌醇与5-Fu联用对肝癌HepG2细胞的增殖抑制和凋亡具有增强作用。
Objective To study the influence of Allopurinol combined with 5-Fluorouracil on the proliferation and apoptosis of liver cancer HepG2 ceils. Methods The HepG2 cells were treated by different concentrations and combinations of Allopwrind, 5- Fluo- rouracil. CCK- 8 was used to measure HepG2 cells proliferation, and flow cytometry was used to examine the apoptosis of HepG2 cells. Results The inhibition of HepG2 cells apoptosis reacted by Allopurinol was weak, in combination inhibited the proliferation of HepG2 cells in a time (24,48,72 h) and concentration dependent (1-3 μg/L) manner. Allopurinol combined with 5-Fluorouracil had stronger inhibitory effect than that was used alone. The apoptosis rate of HepG2 cells in the combination treatment group was 39. 17%, which was significantly higher than 30.86% in the 5-Fluorouracil group(P 〈 0. 05). Conclusion Allopurinol combined with 5- Fluorouracil has enhanced effect on the proliferation and apoptosis of HepG2 cells.
