探讨hsa-miR-199a-5p表达对缺血性脑卒中的预测作用

Expression of hsa-miR-199a-5p in Ischemic Stroke Patients

目的:探讨hsa-miR-199a-5p在缺血性脑卒中(IS)患者血清中的表达和意义。方法:收集118例IS患者及125例健康人群的血清标本。通过实时荧光定量PCR(q PCR)检测血清hsa-miR-199a-5p的表达,用ROC曲线分析hsa-miR-199a-5p表达应用于诊断IS的价值,并对hsa-miR-199a-5p表达与研究人群的临床特征进行相关性分析。结果:hsa-miR-199a-5p在IS患者中的表达显著高于健康人群(P<0.05),且IS患者在疾病的急性期、亚急性期和恢复期时hsa-miR-199a-5p的表达表现出显著的降低趋势(P<0.05)。hsa-miR-199a-5p的表达与IS患者血清CRP水平呈显著正相关(r=0.782,P<0.05)。ROC预测结果显示,hsa-miR-199a-5p诊断IS的灵敏度为94.8%,特异度为96.2%,曲线下面积(AUC)为0.812(95%CI为0.613~0.919,P=0.003)。结论:hsa-miR-199a-5p或许可成为IS的生物标志物。

Objective： To study the expression ofhsa-miR-199a-5p in patients with ischemic stroke （IS）. Meth- ods： The serums of 118 IS patients and 125 healthy people at our hospital were enrolled. The expression of hsa-miR-199a-5p was detected by real-time quantitative PCR （qPCR）. The sensitivity and the specificity were evaluated by the receiver-operating characteristic （ROC） curve and relationship between the expression level of hsa-miR-199a-5p and clinical features was analyzed, respectively. Results： The expression of hsa-miR-199a-5p in the IS patients was significantly higher than that in the healthy control （P〈0.05）. The expression levels of hsa-miR-199a-5p in serum varied at different stages in the IS patients with the lowest expression at the recover stage and highest expression at the acute stage （P〈0.05）. Spearman correlation analysis showed that there was a positive correlation between hsa-miR-199a-5p expression and the serum CRP levels （r=0.782, P〈0.05）. ROC pre- dictions showed that the sensitivity and specificity of hsa-miR-199a-5p confirmed IS were 94.8% and 96.2%, and area under curve （AUC） was 0.812 （95% CI 0.613-0.919, P=-0.003）. Conclusion： Hsa-miR-199a-5p may be used as diagnostic biomarker in IS.
