基于PTEN-PI3K-AKT信号通路探讨非小细胞肺癌Treg细胞上调的分子机制 被引量:6

摘要 目的:通过观察肺癌患者Treg细胞PTEN-PI3K-AKT信号通路分子的表达变化,探讨肺癌患者Treg细胞上调的作用机制。方法:选择30例非小细胞肺癌为研究组,30例健康人作为对照组,采用流式细胞术检测各组人群外周血Treg细胞表达变化;运用免疫磁珠法分选外周血单个核细胞中Treg细胞,Western-blot、Real-time PCR技术检测Treg细胞PI3K、AKT、PTEN蛋白及PTEN mRNA、PI3K mRNA、AKT mRNA的表达情况。结果:研究组外周血Treg细胞占CD+4T细胞的比例(Treg/CD4=4.53%±1.85%)明显高于对照组(Treg/CD4=1.87%±0.93%),差异有统计学意义(P<0.05);与对照组比较,研究组PI3K、AKT蛋白表达明显升高,PTEN表达明显降低,差异有统计学意义(P<0.05);与对照组比较,研究组PI3K mRNA、AKT mRNA表达明显升高,PTEN mRNA表达明显降低,差异有统计学意义(P<0.05)。结论:肺癌患者外周血Treg细胞水平明显升高,其机制可能与PTEN表达下调进而激活PI3K-AKT信号通路相关。 Objective:To observe the expression of PTEN-PI3K-AKT signaling pathway in Treg cells of Lung cancer patients and to explore the mechanism of Treg cells up-regulation in them.Methods:30 cases of non-small cell Lung cancer were selected as the study group and 30 healthy subjects were selected as the control group.The expression of Treg cells in their peripheral blood was detected with flow cytometry.The peripheral blood mononuclear cells were sorted by immunomagnetic beads.The expressions of PI3K,AKT,PTEN and PTEN mRNA,PI3K mRNA and AKT mRNA in Treg cells were detected by Western blot and Western blotting.Results:The percentage of CD4^+T cells in the peripheral blood Treg cells(Treg/CD4=4.53%±1.85%)of the study was significantly higher than that(Treg/CD4=1.87%±0.93%)of the control group(P〈0.05).Compared with that of the control group,the expression of PI3K and AKT protein of the study group were significantly higher respectively while the expression of PTEN was significantly lower(P〈0.05).Compared with that of the control group,the expression of PI3K mRNA and AKT mR- NA were significantly higher respectively while that of the PTEN mRNA were significantly lower(P〈0.05).Conclusion:The level of Treg cell in peripheral blood of patients with Lung cancer is significantly increased,the mechanism of which may be related to the activation of PI3K-AKT signaling pathway.
