夏杞软胶囊对2型糖尿病模型小鼠胰岛素抵抗的影响 被引量:1

摘要 目的:研究夏杞软胶囊治疗2型糖尿病的作用和机制。方法:60只雄性2型糖尿病小鼠随机分为模型组,夏杞软胶囊200 mg·(kg·d)^(-1)组、400 mg·(kg·d)^(-1)组,另取20只雄性小鼠作为对照组,模型组及对照组小鼠给予等量生理盐水。给药6周后进行葡萄糖耐量实验;取血检测空腹血糖、三酰甘油、总胆固醇及空腹血胰岛素,行HE染色进行病理学检查,取肝组织进行Western blotting检测Caveolin-1、胰岛素受体β(insulin receptorβ,IRβ)、p IRβ、蛋白激酶(protein kinase,AKT)、p AKT及细胞膜葡萄糖转运体2(glucose transporter 2,GLUT2)水平。结果:与模型组相比,夏杞软胶囊200 mg·(kg·d)^(-1)组、400 mg·(kg·d)^(-1)组空腹血糖、三酰甘油、总胆固醇水平及HOMA-IR指数显著降低,空腹血胰岛素水平显著升高;肝组织Cavealin-1蛋白表达显著降低;肝组织IRβ,AKT磷酸化水平及GLUT2膜转位显著增加。结论:夏杞软胶囊可降低2型糖尿病小鼠血糖血脂水平,降低肝组织Cavealin-1蛋白表达,增加肝组织IRβ、AKT磷酸化水平及GLUT2膜转位,以改善胰岛素抵抗。 Objective: To study the effect and mechanism of Xiaqi Soft Capsule on the Insulin Resistance of type 2 diabetes model mice. Methods: Sixty male mice were randomly divided into model group,Xiaqi Soft Capsule 200 mg·(kg·d)^-1 group and Xiaqi Soft Capsule 400 mg·(kg·d)^-1 group. Another 20 male mice were used as the control group. The model group and the control group were given the same amount of saline. 6 weeks later,the administration of glucose tolerance test was done. The blood glucose fasting glucose,triglyceride,total cholesterol and fasting blood insulin were detected with HE staining assay,and the liver tissue was detected with Western blotting assay. Caveolin-1,IRβ,p IRβ,Protein kinase( AKT),p AKT and cell membrane glucose transporter 2 antibody( GLUT2) levels were detected. Results: Compared with that of the model group,the fasting blood glucose,triglyceride,total cholesterol and HOMA-IR index were significantly lowered in 200 mg·(kg·d)^-1 group and 400 mg·(kg·d)^-1 group and the fasting blood insulin levels significantly increased. The expression of Cavealin-1 protein in liver tissue significantly decreased,and the levels of IRβ,AKT phosphorylation and translocation of GLUT2 membrane significantly increased in liver tissue.Conclusion: Xiaqi Soft Capsule can reduce blood glucose and blood lipid levels in type 2 diabetic model mice,by decreasing the expression of Cavealin-1 protein in liver tissue and increasing the level of IRβ,AKT phosphorylation and translocation of GLUT2 membrane in liver tissue to improve insulin resistance.
