摘要
用Triton X-100等四种表面活性剂处理牛心包组织,对处理液作蛋白聚糖定量分析,对处理后的组织作蛋白聚糖超微形态观察,并将处理后的材料植入动物皮下21天,通过材料钙含量分析判断钙化程度。结果表明,牛心包组织的钙化与其蛋白聚糖有关。
Bovine pericardium (BP) was treated by nonionic (Triton X-100, Tween 80), anionic (SDS) and cationic surfactants(Hexadecyltrimethy ammonium bromide, HTAB) and concentrations of proteoglycans (PG) of the four different surfactant tanned solutions were quantitated by Bitter's method. Some of the treated BP tissues were morphologically checked for PG in ruthenium red stained sections under transmission electron microscope; some were subdermally implanted in the back of rabbits (New Zealand female albiao rabbites, 8 to 9 weeks of age) for 21 days. Ca~(2+) contents of the BP tissues were quantitatively analyzed. The results indicated that PG contents in Triton X-100, Tween 80 and SDS tanned solutions were three to six times as much as that in HTAB. Under electron microscope, PG granules in BP tissues tanned by the former three surfactants were markedly de creased in comparing with that by the latter one, while the Ca~(2+) contents of the former threa corresponding implanted BP tissues were only 1/30 of that tanned by HTAB, no marked diffarence was found between HTAB and control group.It indicates that calcification of bovine pericardial tissue might be related to its PG content.
