摘要
本研究对水稻铜/锌超氧化物歧化酶铜伴侣(OsCCS)的基本特征、分布情况及功能做了初步探究。通过同源蛋白序列比对,发现OsCCS在多个物种中保守存在。通过芯片分析,结果表明OsCCS热胁迫后表达上调,且OsCCS在各组织中表达有差异,其中在叶中的表达量高,在茎、穗中表达量低。通过实时定量PCR对芯片结果进行验证,发现与芯片结果一致。构建了pCAMBIA2300-OsCCS-GFP载体,通过转化烟草瞬时表达,发现OsCCS蛋白定位于细胞核。上述研究为深入了解OsCCS在水稻中的生物学功能提供了基础。
In this paper, the basic characteristics, distribution, and functions of copper chaperone for Cu/Zn superoxide dismutase (OsCCS) in rice were preliminarily studied. By homologous protein sequence alignment, we found that OsCCS had highly conserved sequence in multiple species. The results of microarray analysis showed that the expression of OsCCS was up-regulated after heat stress, and its expression was variant in different tissues. Among those, OsCCS had the highest expression in the leaves, but the lowest in the stem and spike. The results of microarray verification by real-time quantitative PCR were found to be consistent with the microarray results. In addition, the pCAMBIA2300-OsCCS-GFP vector had been constructed for transient expression in tabacoo, the results of which demonstrated that the OsCCS protein was located at the nucleus. These studies might lay the foundation for the profound understanding of the biological function of OsCCS in rice.
