王不留行对糖尿病胃轻瘫大鼠胃窦平滑肌自发运动的影响

Effects of Wangbuliuxing(Semen Vaccariae) on the Spontaneous Motion of the Gastric Antral Smooth Muscle in Diabetic Gastroparesis Rats

目的:观察王不留行[Vaccaria segetalis(Neck.)Garcke,VSG]对糖尿病胃轻瘫(diabetic gastroparesis,DGP)大鼠胃窦平滑肌自发运动的影响。方法:(1)用链脲佐菌素建造糖尿病大鼠模型;(2)测定胃内色素残留率和小肠传输速率确立胃轻瘫模型;(3)用恒温灌流浴槽,将离体肌条分为正常组和DGP组,DGP组又分为:VSG组、VSG+异搏定(10-7mg·L^(-1))组、VSG+酚妥拉明(10-6mg·L^(-1))组、VSG+苯海拉明组(10-6mg·L^(-1))、VSG+消炎痛组(10-6mg·L^(-1))和VSG+阿托品组(10-6mg·L^(-1))6组,以探讨VSG对DGP大鼠胃窦离体肌条自发收缩的影响及机制。结果:VSG能增强DGP大鼠胃窦离体肌条收缩的振幅、持续时间和收缩面积,但对频率(DGP组:4.50±0.83,VSG组:4.51±0.46)无显著影响;而异搏定和阿托品能明显抑制VSG增强平滑肌活动的效应;酚妥拉明、苯海拉明和消炎痛则无影响。结论:VSG增强DGP胃窦平滑肌的收缩可能与细胞膜上M受体和钙通道有关,与α受体、H受体和前列腺素受体无关。

To observe the effects of Wangbutiuxing （Semen Vaccariae） on the spontaneous motion of the gastric antral smooth muscle in diabetic gastroparesis rats. Methods：Model rats with diabetes were made with streptozotocin（STZ） ;the pigment residual rate in the stomach and the transmission rate of small intestine were determined to establish gastroparesis models ; perfusion the bath machine under constant temperature was performed and the muscle strips in vitro were divided into normal group and DGP group,and DGP group were further divided into. VSG group, VSG ＋ verapamil group （10-7 m· L-1）, VSG ＋ phentolamine group （10-6 mg · L-1） ,VSG ＋ benzhydramine group （ 10 -6 mg · L-1 ） ,VSG ＋ indomethacin group （ 10 -6 mg - L-1） ,VSG ＋ atropine group （10-6 mg ·L -1 ）. The influence and mechanism of VSG on the spontaneous contraction effects of gastric antral strips in vitro were explored. Resuhs：VSG can increase the amplitude, duration and contraction area of the strips in vitro in DGP rats, but it had no significant influence on the frequency （ model group 4.50 ±0.83, VSG group ：4.51 ± 0.46 ）. Verapamil and atropine can inhibit the VSG, ＇s effects of strengthening the smooth muscle motion;But phetolamine, benzhydramine and indomethacin have no contraction effects. Conclusion：VSG can strengthen the motion of smooth muscles,which may be related to the M receptor on the cytomembrane and calcium channel, but it has no relationship with receptor α, H receptor and prostaglandin reeeptor.
