芹菜素对肺癌细胞的抗增殖作用和抗肿瘤药物的增敏作用 被引量:6

Anti-Proliferative Effect of Apigenin on Lung Cancer Cells and Its Sensitization Effect of Anti-Tumor Drugs

摘要 目的探讨芹菜素对肺癌细胞的抗增殖作用和抗肿瘤药物的增敏作用。方法选择鼠龄约5周A/J系小鼠140只,随机分成3组,第1组40只,第2组60只,第3组40只,每组均平分成试验组及对照组。鼠龄6~7周时,在第1组中试验组、第2组经腹腔注射4-(甲基亚硝胺基)-1-(3-吡啶)-1-丁酮(NKK)10 mg/kg,第1组中对照组腹腔注射生理盐水;第2~3 d,第2组中试验组每日大剂量芹菜素灌胃;第3组给予NKK腹腔注射,给药第52~62周,试验组每日芹菜素大剂量灌胃;第2组中对照组、第3组中对照组小鼠分别以植物油大剂量灌胃。在试验第52周将第1组、第2组小鼠处死,63周第5 d将第3组小鼠处死;3组小鼠均被处死解剖切片观察,并利用奥沙利铂及博来霉素对肺癌切片进行处理。结果第1组中试验组平均发现(1.35±0.22)个肺癌病灶,对照组未见癌性病灶;第2组中试验组仅2只小鼠偶见肺癌病灶,对照组发现(1.71±1.28)个癌性病灶;第3组中试验组发现(1.01±1.19)个癌性病灶,对照组发现(1.56±0.78)个癌性病灶。抑制率第1组为0,第2组为(94.32±0.27)%,第3组为(0.36±0.03)%;芹菜素与奥沙利铂及博来霉素联用存在一定的协同作用(CI多小于1)。结论芹菜素对肺癌细胞的抗增殖作用和抗肿瘤药物的增敏作用良好。 Objective To analyze the anti-proliferative effect of apigenin on lung cancer cells and its sensitization effect of anti-tumor drugs. Methods Totally 140 A/J mice of 5 weeks were selected and randomly divided into 3 groups,40 mice in the first group, 60 mice in the second group,40 mice in the third group,they were divided into the experimental group and the control group accord-ing to the digital random principle. In the rats aged 6-7 weeks,the experimental group in the first group and the second groups were intraperitoneally injected with 4-(methyl nitrosamine ) -1- (3-pyridine ) -1- butanone(NKK ) 10 mg/kg,and the control group in the first group was intraperitoneally injected with physiological saline. On the second and third day,the experimental group in the second group was treated with large doses of apigenin,the third group was given intraperitoneal injection of NKK. In the 52-62 weeks,the ex-perimental groups were given intragastric administration of apigenin at large doses,the control groups in the second group and the third group were treated with large doses of vegetable oil. In the fifty-second week,the mice in the first group and the second group were killed,and the mice in the third group were killed on the fifth day in the sixty-third weeks. The mice in the 3 groups were sacrificed and dissected to observe,and the lung cancer slices were treated with Oxaliplatin and Bleomycin. Results In the first group,an average of (1. 35 ± 0. 22)lung cancer lesions were found in the experimental group,and there were no cancer lesions in the control group. In the second group,only 2 mice in the experimental group were occasionally found the lung cancer lesions,and (1. 71 ± 1. 28 ) cancer le-sions were found in the control group. In the third group,(1. 01 ± 1. 19 ) cancer lesions were found in the experimental group,and (1. 56 ± 0. 78 ) cancer lesions were found in the control group. The CI were 0,(94. 32 ± 0. 27)% and (0. 36 ± 0. 03)% in the first,second and third group,respectively. Apigenin combined with Oxaliplatin and Bleomycin had synergistic reaction (CI was mostly less than 1 ) . Conclusion Apigenin plays an important role in the anti-proliferation of lung cancer cells and the sensitizing effect of anti-tumor drugs.
