摘要
旨在克隆牦牛KLF2(Krupple-like transcription factor 2,Kruppel样转录因子2)基因功能区序列,阐明其组织表达规律,并揭示基因与肌内脂肪含量的相关性。以麦洼牦牛为试验动物,利用RT-PCR方法克隆KLF2基因功能区序列,荧光定量PCR技术检测该基因在各种组织中的表达模式。结果表明,获得牦牛KLF2基因序列454bp(GenBank登陆号:KX395178),包括KLFs家族典型的3个锌指结构域;KLF2mRNA在牦牛的肺脏和脂肪组织中存在高水平表达,极显著高于其他组织(P<0.01);KLF2 mRNA的表达水平与背最长肌IMF(Intramuscular Fat,肌内脂肪)含量呈正相关(P<0.01,R=0.885)。结果推测KLF2可能作为牦牛肉质性状的候选基因。
The aim of this study was to clone yak KLF2 gene domain, clarify its expression character- ization in different tissues and reveal the correlation between gene expression and IMF (intramuscular fat). The Reverse Transcription PCR (RT:PCR) was used to clone Maiwa yak KLF2 gene domain, the quanti- tative real-time PCR (qPCR) was used to detect the expression level of KLF2 gene in various tissues. The results showed that the sequence of yak KLF2 gene domain was 454 bp (GenBank accession number: KX395178), contained three KLFs family typical zinc finger domains; The mRNA expression level of KLF2 was high in liver and adipose tissue, which is significantly higher than in other tissues (P'〈0.01) The mRNA expression level of KLF2 was significantly correlated with longissimus IMF content (19 0.01, R=0. 885). This study speculated that KLF2 may he a candidate gene of yak meat quality traits.
