纳米二氧化钛对雌性小鼠肾脏的影响 被引量:3

摘要 为研究在亚急性毒性的情况下,纳米二氧化钛(TiO_2)对雌性小鼠肾脏的影响。采用将60只ICR雌性小鼠随机分为对照组和三个剂量组(10、50、100 mg/kg BW)的方法,连续30 d灌胃染毒。小鼠脱颈处死,称量并计算肾脏系数,观察肾组织病理学变化和氧化损伤,并检测肾功能。结果显示,纳米TiO_2染毒导致小鼠肾脏组织出现不同程度炎细胞浸润,间质充血、纤维素样坏死等损伤,且剂量越大,损伤越明显;各剂量组小鼠肾脏系数和肾组织BUN含量与对照组无显著性差异(P>0.05),而肾组织Cr含量明显升高(P<0.05),UA含量随剂量升高逐渐下降,当剂量达到100 mg/kg时与对照组相比差异显著(P<0.05);小鼠肾组织SOD活性随染毒剂量增加呈先升后降的趋势,且50 mg/kg剂量组SOD活性显著性高于对照组(P<0.05);与对照组相比,10 mg/kg剂量组和50 mg/kg剂量组小鼠肾组织GSH-PX活性显著性升高(P<0.05),各剂量组小鼠肾组织MDA含量明显升高,差异具有统计学意义(P<0.01)。由此可知,纳米TiO_2染毒使雌性小鼠肾组织产生病理学变化,造成肾功能损伤,并引起肾组织的氧化应激反应。 The subacute toxic effects of nano titanium dioxide(TiO_2) on the kidney of female mice were studied in this research. 60 female ICR mice were randomly assigned into 10, 50 and 100 mg/kg nano-TiO_2 groups and control group(phosphate buffer solution). After 30 consecutive days of intragastric administration of corresponding nano-TiO_2, the mice were sacrificed. Then the kidney were used for assessing tissue coefficient and pathological changes; and the level of renal functions indices and oxidative stress markers of kidney were analyzed. Histopathologic examination showed there are various damage in the kidneys of mice exposed to nano-TiO_2. There are no significant differences in the renal coefficient and the content of BUN of kidney among the four groups. The Cr content in nano-TiO_2 treated groups were significant increased compared with that in control group(P0.05). The UA content in 100 mg/kg group was significant decreased compared with that in control group(P0.05). With the nano-TiO_2 dosages increased, the activity of SOD and GSH-PX in the kidneys were first increased, and then decreased. Compared with those in control group, the SOD activity in 50 mg/kg dose group and The GSH-PX activity in10mg/kg and 50 mg/kg BW dose groups were increased significantly(P0.05). The MDA content was significantly increased in nano-TiO_2 treated groups compared with that in control group(P0.01). Nano-TiO_2 can induce the change of renal tissue and functions and the oxidative stress in the kidneys.
