ML-超微量磁珠法和Chelex-100法提取脱落细胞的比较研究 被引量:1

摘要 目的对ML-超微量磁珠法(King Fisher Flex)和Chelex-100法提取脱落细胞DNA的检验效果进行比较,为优化案件里脱落细胞检材的提取方法提供方向。方法对同一份检材(纺织品或作案工具)利用两种提取方法进行DNA的平行提取,常规STR检测,经卡方检验比较差异。结果 112份纺织品检材中ML-超微量磁珠法检出率为58.0%,Chelex-100检出率则为45.5%,差异具有统计学意义(χ~2=7.00,P<0.05)。130份作案工具检材中ML-超微量磁珠法检出率为41.5%,Chelex-100检出率则为33.8%,差异具有统计学意义(χ~2=4.55,P=0.03)。结论 ML-超微量磁珠法(King Fisher Flex)提取脱落细胞更具有优势。 Objective To compare the extraction rate of DNA from exfoliated cells by ML-ultra micro beads method (KingFisher Flex) and Chelex-100 method, thus optimizing the extraction method in forensic practice. Method The parallel extraction of DNA from the same sample (textiles or tools) was carried out by two extraction methods, and the results were compared with common STR. Result Among 112 textile samples,the detection rate of ML-micro beads method and Chelex-100 method was 58% and 45.5%, respectively. The difference was statistically significant (x2=7.00, P〈0.05). Among 130 pieces of crime tool samples, the detection rate of ML-micro heads method and Chelex-100 method was 41.5% and 33.8%, respectively. The difference was also statistically significant (X2=4.55, P=0.03). Conclusion ML-micro beads method (KingFisher Flex) has more advantages in the extraction of DNA from exfoliated cells.
