试验采用PHENOMENEX LUNA C18色谱柱(250 mm×4.6 mm,5μm),流动相A相为0.5%乙酸水溶液,流动相B相为80%甲醇水溶液,梯度洗脱,流速为0.8 m L/min,DAD检测波长为285 nm,柱温为30℃。研究比较不同来源辛夷花提取物对原儿茶酸、间羟基苯甲酸、对羟基苯甲酸、对香豆酸、阿魏酸、丁香酸、肉桂酸、芥子酸和绿原酸9种酚酸物质的HPLC检测及辛夷花来源对其酚酸物质含量的影响。检测方法显示:9种酚酸物质的检测线性范围为0.05~200 mg/L;检出限(S/N=3)在0.001~0.05 mg/kg之间,在加标浓度为5.0~20.0 mg/kg条件下,加标回收率(N=9)在95.18%~107.68%之间,精密度为0.98%~3.24%(n=6),检测方法灵敏、有效。检测结果表明,不同来源辛夷花提取物得到的辛夷花提取物中原儿茶酸、间羟基苯甲酸、对羟基苯甲酸、对香豆酸、阿魏酸、丁香酸、肉桂酸、芥子酸和绿原酸9种辛夷花提取物酚酸物质的总含量以陕西渭南产辛夷花提取物最高。
In this study, the influence of different Magnolia liliftora Desr. extracts on contents of nine kinds of the phenolic acids (Protocatechuic acid, m-hydroxybenzoic acid, p-hydroxybenzoic acid, p-coumaric acid, ferulic acid, cinnamic acid, cinnamic acid, sinapic acid and chlorogenic acid) in Magnolia liliflora Desr. extracts was compared by using HPLC. The experiment was based on PHENOMENEX LUNA C18 column (250 mm ×4.6 mm, 5 μm) with mobile phase A of 0.5% acetic acid aqueous solutionand mobile phase B of 80% methanol aqueous solution, and gradient eluted with the flow rate of 0.8 mL/min, DAD detection wavelength of 285 nm and column temperature of 30 ℃. Detection method showed sensitive and effective: ninekinds of phenolic acidslinear ranged of 0.05-200 mg/L; The detection limit (S/N=3) was between 0.001-0.05 mg/kg, precision was 0.98%-3.24% (n=6), the recoveries were between 95.18% and 107.68% in the spiked 5.0-20.0 mg/kg concentration. It was shown by the results that the total content of ninephenolic acids (protocatechuic acid, m-hydroxybenzoic acid, p-hydroxybenzoic acid, p-coumaric acid, ferulic acid, cinnamic acid, cinnamic acid, sinapic acid and chlorogenic acid) in extracts and the highest in Shaanxi Weinan Magnolia liliflora Desr. extracts.
