摘要
目的:经小鼠球后静脉注射氯化钙(CaCl_2)-乙酰胆碱(Ach)混合液,探索一种能诱发小鼠产生心房颤动且造模周期短的心房颤动模型制备方法与技术。方法:将选取健康成年昆明小鼠24只,体质量35~55g,随机分为三组,每组8只,雌雄各半,称质量,腹腔注射10%水合氯醛(3mL/kg)麻醉,待麻醉后,经小鼠球后静脉注射不同浓度的CaCl_2-Ach混合液(内含CaCl_25~10mg/mL,Ach 25~50μg/mL),用量为0.1mL/10g,5s内注射完,多导生理记录仪记录小鼠心电图变化,间隔24h,连续给药7d,观察每次给药后小鼠心房颤动持续时间。结果:经球后静脉注射CaCl_2-Ach混合液后,三组小鼠均可诱发出心房颤动,且雌雄产生心房颤动持续时间差异无统计学意义(P=0.775);给予CaCl_25mg/mL+Ach25μg/mL混合液,第四天起心房颤动持续时间趋于稳定(30.5±3.5)s。结论:经小鼠球后静脉注射CaCl_2-Ach混合液可以诱导产生心房颤动,且造模周期短,持续时间趋于稳定,有利于临床心房颤动机制的研究。
Objective: Mixed Ach-CaCl2 solution was injected into the retro-orhital vein of Kunming mouse at different concentrations, and then recorded the ECG of the mouse, every 24 hours, in 7 days, in order to build a method which could induce atrial fibrillation in mouse. Methods: To build a atrial fibrillation model in mouse, twenty-four adult and health KungMing mouse(35-55g) were recruited, divided randomly into three subgroups, half male and half female, chloral hydrate was injected by intraperitoneal injection in 5 seconds, af- ter anesthetization, mixed Ach-CaC12 solution was injected into the retro-orbital vein of KunMing mouse at dif- ferent concentrations( including CaC12 5 -10mg/mL, Ach 25-50μg/mL) every 24 hours, in 7 days, electrocar- diogram was recorded by BL-420F biological systems everytime before and after the treatment. Results: The mixed solution of Ach-CaCl2 could induce atrial fibrillation in three groups, and there is no difference between male and female mouse (P = 0. 775 ). Especially, the concentration of CaC12 5mg/mL and Ach 25txg/mL could induce more stable ( 30. 5 ± 3.5 ) s duration of atrial fibrillation after the fourth day of the treatment. Conclu- sion : Atrial fibrillation model in KunMing mouse can be induced by retro-orbital vein injection of suitable Ach- CaC12 solution, and it is simple and stable without long time to build the model for researching the mechanism of atrial fibrillation.
