中药抑肺饮对Lewis肺癌移植瘤生长及细胞EMT的影响

Effect of Traditional Chinese Medicine Formula Yifei Yin on the Growth of Lewis Lung Cancer Xenografts and the Regulation of EMT in Lung Cancer Cells

目的:观察中药抑肺饮对C57BL/6小鼠Lewis肺癌移植瘤的抑制作用,研究其对细胞EMT的影响。方法:70只雄性SPF级C57BL/6小鼠随机分组为空白对照组、模型组、顺铂组、抑肺饮联合顺铂组、抑肺饮高剂量组、抑肺饮中剂量组和抑肺饮低剂量组,建立Lewis肺癌移植瘤模型,给药15 d后脱颈处死小鼠,取肿瘤组织并称重,计算各组抑瘤率。对肺组织进行HE染色观察各组小鼠肺组织病理变化情况,同时检测肿瘤组织中E-cadherin和Vimentin的表达情况。结果:抑肺饮具有抗肿瘤作用,其中抑肺饮联合顺铂组的抑瘤率最高,为72.39%。与模型组相比较,抑肺饮作用组瘤组织中的E-cadherin表达量呈上调趋势,Vimentin的表达量呈下调趋势,其中抑肺饮联合顺铂组的差异最为明显。结论:抑肺饮联合顺铂组显著抑制了C57BL/6小鼠Lewis肺癌移植瘤的生长及Vimentin的表达,同时,上调E-cadherin的表达,说明抑肺饮的作用机制可能和调节肿瘤细胞EMT相关。

Objective：To observe the inhibitory effect of traditional Chinese medicine formula Yifei Yin on Lewis Lung cancer xenografts in C57BL/6 mice,and to study its effect on the regulation of EMT in lung cancer cells.Methods：A Lewis lung cancer xenograft model was established in male C57BL/6 mouse.Totally 70 C57BL/6 mice were randomly divided into normal group,model group,DDP group,Yifei Yin combined with DDP group,and high,middle and low dose Yifei Yin groups（n=10 in each group）.After 15 days,the mice were sacrificed by neck luxation and the inhibition rate of Yifei Yin was calculated according to the final tumor weight.Pathological changes were observed by HE staining.The expression levels of E-cadherin and Vimentin were detected by immunohistochemistry and real-time PCR.Results：Yifei Yin can inhibit the growth of lung cancer cells.The inhibition rate of Yifei Yin combined with DDP group（72.39%） was higher than those of other groups.Compared with the model group,the expression level of E-cadherin was increased in the Yifei Yin treatment groups,and the expression level Vimentin were decreased.The most significant difference was observed in the Yifei Yin combined with DDP group.Conclusion：Yifei Yin combined with DDP can inhibit the growth of Lewis lung cancer xenografts in C57BL/6 mice and the expression level of Vimentin,and increases E-cadherin expression.These results suggest that the inhibitory effect of Yifei Yin may be related with suppression of EMT in lung cancer cells.
