马鹿茸不同分子量肽段中水解及游离氨基酸的含量分析 被引量:10

摘要 目的:测定马鹿茸不同分子量肽段中水解氨基酸和游离氨基酸的含量及构成。方法:采用柱前衍生化,氨基酸自动分析仪对马鹿茸肽段中氨基酸类成分进行含量测定。色谱柱为氨基酸分析专用ODS柱,柱温:27℃;检测波长360 nm;流动相总流速1.2 mL·min^(-1)。结果:马鹿茸肽段中氨基酸类成分线性关系良好,r均大于0.999;精密度、稳定性、重复性试验的RSD均小于3%;加样回收率在96.6%~104.7%之间,RSD值均小于2.4%。不同分子量肽段的氨基酸含量及构成有一定差异。50~100 k分子量段的氨基酸含量最高。结论:该方法简便可行、重复性好、准确度高,可用于马鹿茸药材多肽中水解氨基酸及游离氨基酸的含量测定。 Objective:To determine the contents and compositions of hydrolysis and free in different molecular weight velvet antler peptide segment.Method:With the method of pre-column derivatization,determine the contents of amino acids in velvet antler peptide segment by amino acid analyzer.The column was dedicated for determinating amino acids,ODS column.The flow rate was 1.2 mL·min-1,wavelength was set at 360 nm and column temperature was at 27 ℃.To determine the contents and compositions of hydrolysis and free in different molecular weight velvet antler peptide segment.Result:The results were the correlation coefficients of amino acids in velvet antler peptide segment were all greater than 0.997.The precision,stability and repeatability of RSD were less than 3% and the recovery was all between 96.6%104.7% with RSD less than 2.4%.The contents and compositions had some differences in different molecular weightamino acids,50100 k was the highest.Conclusion:We have agreements that the method is simple,reproducible and accurate.It can be used for determinating the contents of hydrolysis and free amino acids in velvet antler peptide segment.
