摘要
目的:探讨脾阳虚模型大鼠"运""化"功能的增龄性变化。方法:将新出生40只Wistar大鼠,体质量(5.6±0.5)g,采用肩胛骨间棕色脂肪组织切除术及高脂饮食隔日寒冷环境刺激造模,术后随机分为8周龄模型组、16周龄模型组、32周龄模型组、64周龄模型组4组,每组10只。另外40只大鼠用来设立相对应的同周龄的对照组4组,每组10只。到了相应的周龄后,检测胃肠动力学指标,采用免疫组化检测胃促生长素表达,酶联免疫吸附法和放射免疫法检测血清胃促生长素、胃泌素、瘦素、神经肽Y水平。结果:32周龄模型组、64周龄模型组分别与同周龄对照组比较,在胃肠动力学指标差异有统计学意义(P<0.05或P<0.01),血清胃泌素、瘦素、神经肽Y水平差异也均有统计学意义(P<0.05或P<0.01),免疫组化检测结果显示胃促生长素表达有增龄性递减趋势。结论:脾阳虚模型大鼠"运""化"功能随增龄减弱,与胃促生长素调节胃肠运动和调控胃泌素、瘦素、神经肽Y水平通路机制有关。
Objective:This study was aimed to investigate the age-related changes of functions of transportation and transformation in rats with spleen yang deficiency syndrome.Methods:Forty new born Wistar rats,weighted(5.6±0.5)g with brown adipose tissue resection,were given high fatty feeding and cold environment stimulating every other day to set up model and then were divided into 8 weeks,16 weeks,32 weeks and 64 weeks groups with 10 in each group.In addition,other 40 rats were as control groups with the same and corresponding age groups for 4 groups with 10 in each group either.Then detect the indexes of gastrointestinal motility and the serum levels of ghrelin,gastrin(GAS),leptin(Lp),neuropeptide Y(NPY) with ELISA and radio-immunity and observe the expression of ghrelin with immunohistochemical(IHC) technolygy.Results:The results showed that compared with the same age control group,in 32 weeks and 64 weeks groups,there was statistical significance about the indexes of gastrointestinal motility(P〈0.05 or P〈0.01).Moreover,there was also statistical significance about the serum levels of ghrelin,GAS,Lp or NPY(P〈0.05 or P〈0.01).Meantime,the result of IHC showed that the expression of ghrelin reduced with the age increasing.Conclusion:It was concluded that functions of transportation and transformation in rats with spleen yang deficiency syndrome got weaken,which might be involved with the mechanism of ghrelin regulating gastrointestinal motility and the levels of GAS,Lp and NPY.
