摘要
目的建立HPLC法同时测定人工养殖复齿鼯鼠所产五灵脂中槲皮苷、杨梅苷2种成分的含量。方法采用Diamonsil ODS C_(18)色谱柱(4.6mm×250mm,5μm),流动相:甲醇-0.01mol·L^(-1)磷酸二氢钾溶液-冰醋酸(28∶72∶1.5),流速:1.0ml·min^(-1),检测波长:262nm,柱温:40℃。结果槲皮苷的线性范围为0.0411~0.8216μg(r=0.9996),平均回收率99.17%(n=6,RSD=1.58%);杨梅苷的线性范围为0.0181~0.3616μg(r=0.9997),平均回收率98.88%(n=6,RSD=1.76%)。结论该方法可作为人工养殖复齿鼯鼠所产五灵脂药材中槲皮苷、杨梅苷等2种成分含量测定的方法。
Objective To establish an HPLC method for simultaneous determination of quercitroside and myricetrin in excrement of breeding state trogopterus xanthipes.Methods The HPLC system consisted of Diamonsil ODS C18 column(4.6 min×250 mm,5 μm) with a solution system of methanol-0.01 mol ·L^-1 KH2PO4-glacial acetic acidsolution (28 : 72 : 1.5) as the mobile phase.The flow rate was 1.0 ml ·min^-1 , and the detection wavelength was 262 nm at a column temperature of 40 ℃.Results The linear range of quercitroside was 0.0411 -- 0. 8216 μg ( r = 0. 9996), and the average recovery was 99.17 % ( n = 6, RSD = 1.58%).The linear range of myricetrin was 0.0181--0.3616 μg( r =0.9997),and the average recovery was 98.88%(n :6, RSD= 1.76 %).Conclusion This method can be used for the determination of two compo- nents in excrement of breeding state trogopterus xanthipes.
