摘要
目的:观察艾炷灸对D-半乳糖衰老小鼠学习记忆及血清、脑组织胰岛素(INS)水平的影响。方法:将75只小鼠随机分为对照组、造模组、艾灸1组、艾灸2组、艾灸3组,每组15只。艾灸1组艾炷灸足三里穴+悬钟穴,艾灸2组艾炷灸百会穴+关元穴,艾灸3组艾炷灸腕背关节上1mm与膝髌正中上沿上2mm。用Morris水迷宫测试学习记忆力,用酶联免疫法检测血清及脑组织INS水平。结果:治疗前各组小鼠水迷宫各项指标及学习记忆能力比较,差异无统计学意义(P>0.05);治疗后,造模组与对照组、艾灸1组、艾灸2组比较,各项指标差异均有统计学意义(P<0.01或P<0.05);对照组、艾灸1组、艾灸2组与艾灸3组比较,差异均有统计学意义(P<0.05)。造模组血清和脑组织INS水平较对照组、艾灸1组和艾灸2组升高或降低,差异均有统计学意义(P<0.01或P<0.05);对照组、艾灸1组、艾灸2组与艾灸3组比较,差异均有统计学意义(P<0.05)。结论:艾炷灸足三里穴+悬钟穴、关元穴+百会穴均能显著提高D-半乳糖衰老小鼠学习记忆能力,与降低外周循环胰岛素水平、提高脑组织的胰岛素浓度有关,具有延缓脑衰老作用。
Objective: To investigate the effect of moxa cone moxibustion on learning and memory abilities and insulin( INS) level in serum and cerebral tissue in mice with senescence induced by D-galactose. Methods: A total of 75 mice were randomly divided into control group,model group,and moxibustion groups 1,2,and 3,with 15 mice in each group. The mice in moxibustion group 1 were given moxa cone moxibustion at Zusanli and Xuanzhong points,those in moxibustion group 2 were given moxa cone moxibustion at Baihui and Guanyuan points,and those in moxibustion group 3 were given moxa cone moxibustion at 1mm above the waist back joint and 2mm above the upper edge of the middle knee-patellar joint. The Morris water maze was used to test learning and memory abilities,and ELISA was used to measure INS level in serum and cerebral tissue. Results: There were no significant differences in the indices of water maze test and learning and memory abilities between groups before treatment( P〉0. 05); after treatment,there were significant differences in these indices between the model group and the control group/moxibustion groups 1 and 2( P〈0. 01 or P〈0. 05),as well as between the control group/moxibustion groups 1 and 2 and moxibustion group 3( P〈0. 05). Compared with the control group and moxibustion groups 1 and 2,the model group had a significant increase or reduction in the INS level in serum and cerebral tissue( P〈0. 01 or P〈0. 05),and there was a significant difference between the control group/moxibustion groups 1 and 2 and moxibustion group 3( P〈0. 05). Conclusion: Moxa cone moxibustion at Zusanli and Xuanzhong points or Baihui and Guanyuan points can significantly improve the learning and memory abilities of mice with senescence induced by D-galactose,possibly by reducing INS level in peripheral circulation and increasing INS level in cerebral tissue,and therefore,it can delay brain aging.
