摘要
为研究禽呼肠孤病毒(ARV)在鸡肝癌细胞(LMH)中的增殖特性,本研究将ARV S1133毒株感染LMH细胞,连续传代3次,观察细胞病变(CPE),通过RT-PCR鉴定病毒,并进一步研究病毒的增殖特性。结果显示,ARV S1133毒株能够在LMH细胞中很好地增殖,并出现典型的CPE。一步生长曲线结果显示,S1133感染LMH细胞后的前12 h为潜伏期,12 h^24 h为快速增长期,第24小时的效价达到最大,TCID50为10-9.2/0.1 m L。本研究为进一步研究ARV的致病机理和研制细胞灭活疫苗提供了良好的操作平台。
To study the proliferation characteristics of avian reovirus(ARV) in chicken hepatocarcinoma cells(LMH),LMH cells was infected with ARV S1133 strain and continuously passaged for three times.The cytopathic effect(CPE) was observed after infection,and the proliferation characteristics of the virus was studied by the Reed-Meunch method.The results showed that ARV S1133 strain was well-valuable in LMH cells and showed typical CPE.The results of one-step growth curve showed that S1133 was incubated for 12 h after infected with LMH cells,rapidly grown between 12 h and 24 h and the titer was the highest at 24 h,and TCID50 was 10-9.2/0.1 m L.This study provides a good platform for further studies on the pathogenesis of ARV and the development of cell inactivated vaccines.
