诱导型腹水综合征肉鸡心肌的病理变化与增殖

Myocardial pathological changes and proliferation in broilers with induced ascites syndrome

为探索诱导型腹水综合征肉鸡心脏的病理变化与心肌细胞增殖之间的关系,将20日龄白羽肉鸡分成对照组(静脉注射肝素钠生理盐水,n=30)和试验组(静脉注射含纤维素微粒的肝素钠生理盐水,n=90)。自注射纤维素微粒后,记录腹水综合征(AS)发病率,并于肉鸡42日龄时取样,测定右心室重量与全心室重量之比(mrv/mtv)、红细胞压积(PCV)以及血清中谷草转氨酶(AST)活性浓度和肌酸激酶(CK)活性浓度;右心室组织常规苏木精—伊红染色,并运用实时荧光定量PCR测定右心室组织PCNA和Ki-67的表达量。结果,与对照组相比,试验组肉鸡AS发病率、mrv/mtv值、PCV及血清中AST活性浓度和CK活性浓度显著增高(P<0.01);试验组肉鸡的右心室明显扩张,心肌细胞出现水肿现象,但PCNA和Ki-67的m RNA表达量无明显变化。上述结果表明,诱导型腹水综合征肉鸡心脏产生了严重的病理变化,但心肌所产生的这种病变与细胞的增殖无关。

In order to explore the relationship between cardiac pathological changes and proliferation in the induced ascites syndrome,the 20 days-old broilers were divided into controlgroup（intravenous injection of heparin saline,n =30） and test group（intravenous injection of heparin saline containing cellulose particles,n=90）.The incidence of ascites syndrome（AS） was recorded after the injection of cellulose particles.The ratio between right ventricle and total ventricle（mrv/mtv） in 42days-old chicken was calculated.And the packed cell volume（PCV） was tested.Then the specific activities of the serum aspartate aminotransferase（AST） and creatine kinase（CK） were measured by ext racting the venous blood.Right ventricular tissue was stained routinely by hematoxylin and eosin.The expression of proliferating cell nuclear antigen（PCNA） and the marker of proliferation Ki-67 m RNA wasanalyzed by real time fluorescence quantitative PCR in the right ventricular tissue.In result,the incidence of AS,the mrv/mtvand PCV in the test group were significant higher（P〈0.01） than that of the control group,and so were the ecific activities of the serum AST and CK.Right ventricular tissue was dilatant and cardiac muscle cells were dropsical in broiler chicken of test group.But the expression of PCNA and Ki-67 of m RNA in test group showed no significant difference in comparison with control group.These results indicated that the induced ascites syndrome had severe damages to the heart of broilerchicken,but it was not related to the proliferation of the cells.
