不同剂量黄芪注射液对病毒性心肌炎小鼠Caspase-3活性的影响

Effect of different dose of astragalus injection on the Caspase-3 activity of mice with viral myocarditis

目的观察黄芪注射液对病毒性心肌炎小鼠发生细胞凋亡及对半胱天冬氨酸蛋白酶3(Caspase-3)活性的影响。方法 4~6周龄雄性Balb/c小鼠60只,随机分为正常对照组、模型对照组、黄芪低剂量组、黄芪中剂量组、黄芪高剂量组5组,每组各12只。制备小鼠急性病毒性心肌炎模型后黄芪低、中、高剂量组分别腹腔注射黄芪注射液0.1、0.2、0.4mL/(g·d),正常对照组及模型对照组分别给予腹腔注射生理盐水0.1mL/(g·d),每日1次,连用14d。在第3、7、10、14天取心脏及血清标本,进行病理学检查、Caspase-3表达活性的检测、肌酸肌酶同工酶(CK-MB)及心肌肌钙蛋白Ⅰ(cTnⅠ)的检测。结果 (1)模型对照组小鼠心肌病理积分、CK-MB、cTnⅠ值显著高于正常对照组,差异有统计学意义(P<0.05);(2)黄芪低、中、高剂量组Caspase-3表达活性、CK-MB、cTnⅠ值、心肌病理积分均明显低于模型对照组,差异有统计学意义(P<0.05);(3)黄芪低、中、高剂量组各组间心肌病理积分、CK-MB及Caspase-3表达活性差异无统计学意义(P>0.05)。结论 (1)细胞凋亡参与了病毒性心肌炎发生时小鼠的心肌损伤,Caspase-3在其中起到重要作用;(2)小剂量的黄芪注射液即可以通过下调心肌细胞Caspase-3活性减少病毒性心肌炎小鼠心肌细胞凋亡,减轻心肌细胞损伤。

Objective To observe the effect of astragalus injection on cell apoptosis in mice with viral myocarditis and on the Caspase-3 activity. Methods A total of 60 male Balb/c mice（4 to 6 weeks old） were randomly divided into 5 groups： normal control group （A） , model control group （B ） , low-dose astragalus group（C） , middle-dose astragalus group（D） and high-dose astragalus group（E） ,12 mice in each group. Af-ter the mice model of acute viral myocarditis was made, astragalus injection was injected into the abdominal cavity of the mice in group C,D and E[0. 1,0. 2,0. 4 mL/（g · d ）] , while normal saline was injected in group A and B[0. 1 mL/（g·d）] ,once a day for continuously 14 days. On day 3,7,10, and 14, heart and serum samples were taken for pathological testing and the detection of Caspase-3 activity, CK-MB and cTnI. Re-sults （1）Myocardial pathological score, and the value of CK-MB and cTnl of mice in group B were significantly higher than those of group A, the difference being statistical（P〈CO. 05）. （2）The Caspase-3 activity, CK-MB, cTnl value and myocardial pathological score in group C？D and E were significantly lower than those in group B, with significant difference（P〈0. 05）. （3）There was no statistical difference on myocardial pathological score, CK-MB or Caspas^3 activity among group C,D and E（P〉0. 05）. Conclusion （1）Cell apoptosis is involved in the myocardial damage of mice with viral myocarditis, and Caspas-3 plays an important role in it. （2）Small-dose astragalus injection can reduce the apoptosis of myocardial cells in mice with viral myocarditis by down-regulating the Caspase-3 activity of myocardial cells, and thus alleviate the damage to myocardial cells.
