维药神香草对治疗哮喘大鼠的免疫机制研究 被引量:2

摘要 目的观察维药神香草对哮喘大鼠白介素(interleukin,IL)-6、嗜酸性粒细胞(eosinophil,EOS)的影响,探讨其治疗哮喘的部分免疫机制。方法将健康Wistar大鼠50只随机分为正常对照组、哮喘模型组、地塞米松组、神香草多糖低剂量组、神香草多糖高剂量组5组,每组10只。正常对照组腹腔注射1 mL无菌生理盐水;其余4组激发致敏引喘,每天灌胃给药1 mL(地塞米松组给药为0.5 mg/kg,神香草多糖低、高剂量组灌胃分别为0.65、2.60 g/kg)。药物干预1周后,末次激发结束后24 h内,每组随机取8只大鼠,支气管肺泡灌洗,收取支气管肺泡灌洗液,测量EOS。采用免疫组织化学法检测肺组织中IL-6的表达水平。用图像ProPlus分析软件进行灰度分析。结果 5组间支气管肺泡灌洗液中EOS差异有统计学意义(P<0.001)。哮喘模型组EOS高于正常对照组,差异有统计学意义(P<0.001)。神香草多糖低、高剂量组EOS明显低于哮喘模型组,差异均有统计学意义(P<0.001);神香草多糖低剂量组EOS高于地塞米松组,差异有统计学意义(P<0.01);神香草多糖高剂量组EOS与地塞米松组比较,差异无统计学意义(P>0.05)。IL-6在胞浆表达,支气管纤毛柱状上皮细胞和浸润的炎细胞出现阳性表达,肺泡间质也出现少量的表达。哮喘模型组IL-6的表达高于正常对照组,差异有统计学意义(P<0.001);神香草多糖低、高剂量组IL-6的表达低于哮喘模型组,差异均有统计学意义(P<0.001);神香草多糖低、高剂量组IL-6与地塞米松组比较,差异均无统计学意义(P>0.05)。结论神香草多糖可抑制哮喘大鼠IL-6的分泌,降低肺泡灌洗液中EOS,减轻哮喘大鼠的炎症反应,提示神香草多糖具有较好的抗炎作用。 Objective To observe the effects of Uygur medicine Hyssopus officinalis on interleukin-6(IL-6) and eosinophil(EOS) in asthmatic rats and to explore the mechanism of partial immunization of asthma.Methods Fifty healthy Wistar rats were randomly divided into normal control group,asthmatic model group,dexamethasone group,low dose Hyssopus officinalis polysaccharide group and high dose Hyssopus officinalis polysaccharide group.The negative normal control group was intraperitoneally injected with 1 mL sterile saline,the other four groups were sensitized and induced asthma,the rats in treatment group were administered 1 mL per day(dexamethasone group with the concentration of 0.5 mg/kg,low dose Hyssopus officinalis polysaccharide and high dose Hyssopus officinalis polysaccharide group with 0.65 g/kg and 2.60 g/kg,resectively.After 1 week,at the end of excitation,each group were randomly selected 8 rats,bronchoalveolar lavage fluid(BALF) was collected for EOS.The expression of IL-6 in lung tissues were measured by immunohistochemistry.Results The EOS of BALF was statistically significant(P〈0.001).EOS in asthmatic model group was higher than that in the normal control group,the difference was statistically significant(P〈0.001).EOS counts in the low and high dose groups were significantly lower than those in the asthmatic model group(P〈0.001).EOS in low dose Hyssopus officinalis polysaccharide group was higher than that in dexamethasone group,the difference was statistically significant(P〈0.01).There was no significant difference in the EOS counts between the high dose group and the dexame-thasone group(P〈0.05).The expression of IL-6 in cytoplasm,positive expression of IL-6located in bronchial ciliated columnar epithelial cells and infiltrating inflammatory cells.The expression of IL-6 in the lung tissue of the asthma group was higher than that in the normal control group,the difference was statistically significant(P〈0.001).The expression of IL-6 in low dose group and high dose Hyssopus officinalis polysaccharide group was lower than that in asthmatic model group(P〈0.001).The level of IL-6 in the high and low dose group was slightly higher than that in the dexamethasone group,but the difference was not statistically significant(P〈0.05).Conclusion The Hyssopus officinalis polysaccharide can inhibit the secretion of IL-6 and decrease the EOS content in BALF and relieve the inflammatory reaction in asthmatic rat.The polysaccharide of Hyssopus officinalis may have a good anti-inflammatory effect.
