摘要 目的:采用Box-Behnken效应面法,优选闪式提取金银花中有效成分的工艺条件。方法:采用闪式提取法,以乙醇浓度、料液比和提取时间为主要影响因素,以绿原酸和木犀草苷转移率的总评"归一值"为评价指标进行Box-Behnken试验设计,并用多元线性回归及二项式拟合,建立总评归一值与自变量之间的数学关系,用效应面法预测最佳工艺条件。结果:采用闪式提取,乙醇浓度为66.92%,料液比为18.82,提取时间为1.20 min,提取次数为一次。以此工艺条件提取金银花中绿原酸的转移率为92.87%,木犀草苷的得率为87.55%。结论:优选的提取工艺简单、经济实用,能为金银花药材中有效成分的快速提取及质量控制提供新思路。 Objective: To optimize the flash extraction process of the active ingredients from Lonicerajaponica by Box-Behnken de- sign and response surface methodology, nethodsl Flash extraction was used. With the concentration of ethanol, solid-liquid ratio and extraction time as the main influencing factors and the overall normalization value of the transfer rates of chorogenic acid and luteoloside as the evaluation index, Box-behnken design was performed to screen out the optimal extraction conditions. The mathematics relation- ship between the overall normalization value and the independent variables was established by multiple linear regression and binomial fitting, and response surface methodology was used to predict the optimal process conditions. Results: The optimal extraction conditions were determined as follows: the ethathol concentration was 66.92%, the solid-liquid ratio was 18.82, the extraction duration was 1.20 min and extracted only once. Under the above conditions, the extraction rate of chloragenic acid and total luteoloside was 92.87% and 87.55%, respectively. Conclusion: The optimized technology is simple, economic and practical,which can provide new ideas for the rapid extraction and quality control of active ingredients from Lonicera japonica.
