摘要
目的:建立托盘根药材中蔷薇酸的高效液相色谱-蒸发光散射检测(HPLC-ELSD)测定方法。方法:采用高效液相色谱法,蒸发光散射检测器。色谱柱:Vision HT C_(18)HL柱(250 mm×4.6 mm,5μm);柱温:35℃;流动相:甲醇-0.1%三氟乙酸(66∶34),流速:0.8 ml·min^(-1);ELSD条件:漂移管温度:90℃,载气流速:2.3 L·min^(-1)。结果:蔷薇酸进样量在0.155~4.346μg范围内呈良好线性关系(r=0.999 9),平均加样回收率为100.4%,RSD=1.88%(n=6)。结论:该方法简便,结果准确,重复性好,可用于托盘根药材中蔷薇酸的含量测定。
Objective: To establish an HPLC-ELSD method for the content determination of rosolic acid in Radix Rubi. Methods: The ELSD-HPLC content determination of rosolic acid was set up using a Vision HT CtsHL column(250 mm ×4.6 mm,5μm), the mobile phase was methanol-0.1% trifluoroacetic acid with a flow rate of 0.8 ml win-l, the column temperature was 35℃, the tem- perature of drift tube heater was 90℃ and the gas was with the flow rate of 2.3 L min -1. Results: The linear range of rosolic acid was 0. 155-4. 346 μg ( r = 0. 999 9). The average recovery was 100.4% with RSD of 1.88% ( n = 6). Conclusion: The method is simple and accurate. It can be used for the quality control of Radix Rubi.
