HPLC法同时测定五酯微丸中五味子酯甲、五味子甲素和安五酯素

Simultaneous Determination of Schisantherin,Deoxyschizandrin and Anwuligan in Wuzhi Pellets by HPLC

目的:建立HPLC法同时测定五酯微丸中南五味子木脂素特征成分五味子酯甲、五味子甲素和安五酯素含量的方法。方法:采用HPLC梯度洗脱同时测定五酯微丸中五味子酯甲、五味子甲素及安五酯素的含量,以月旭Ultimate XB-C_(18)(150 mm×4.6 mm,3μm)为色谱柱,四氢呋喃-水为流动相,梯度洗脱,检测波长为228 nm,柱温:35℃。结果:五味子酯甲在0.040~0.805μg的范围内,线性关系良好(r=0.999 9)。平均回收率为97.0%,RSD=1.67%。五味子甲素在0.092~1.846μg的范围内,线性关系良好(r=0.999 9)。平均回收率为102.5%,RSD=1.49%。安五酯素在0.020~0.998μg的范围内,线性关系良好(r=0.999 9)。平均回收率为95.0%,RSD=1.78%(n=6)。结论:本方法简便、准确、重复性好,可提高五酯微丸的质量标准。

Objective： To establish a method for HPLC determination of schisanthein, deoxyschizandrin and anwuligan in Wuzhi pellets simultaneously and control the quality of Wuzhi pellets. Methods： Schisantherin, deoxyschizandrin and anwuligan were deter- mined by HPLC with a chromatographic column of Welch Ultimate XB-C18 （ 150 mm ×4.6 mm,3μm）, the mobile phase was tetrarn- athylene oxide-water with gradient elution. The detectibn wavelength was 228 nm. The column temperature was 35℃. Results： The lin- ear range of sehisantherin was 0. 04043. 805 μg （ r = 0. 999 9 ） , and the average recovery was 97.0% with RSD of 1.67%. The linear range of deoxychizandrin was 0.092-1. 846μg（ r = 0.999 9）, and the average recovery was 102.5% with RSD of 1.49%. The linear range of anwuligan was 0. 02043. 998 μg（ r = 0.999 9）, and the average recovery was 95.0% with RSD of 1.78% （n = 6） . Conclu- sion： The method is simple, accurate and reproducible. It can improve the quality standard of Wuzhi pellets.
