张家口坝上地区人群环氧合酶-2基因多态性、幽门螺旋杆菌感染与胃癌发生风险的相关性分析 被引量:2

Analysis of COX -2 Genetic Polymorphisms and Helicobacter Pylori Infection in the Incidence of Gastric Cancer among Individuals in Zhangjiakou Area

摘要 目的分析张家口坝上地区人群环氧合酶-2(COX~2)基因多态性、幽门螺旋杆菌(Hp)感染与胃癌发病的关系。方法选取2015年5月~2017年2月于张家口坝上地区居住10年以上的居民200例为研究对象。将研究对象分为胃癌组(n=100)及对照组(n=100)。采用聚合酶链反应-限制性片段长度多态性技术(PCR—RFLP)对两组进行基因分型,以Warhin—starry嗜银法检测两组样本中Hp感染情况。然后经非条件性Logistic回归分析COX-2基因单核苷酸多态性(SNP)rs3218625、Hp感染与胃癌发病风险的相关性。结果Logistic回归分析显示COX-2基因SNP rs3218625与胃癌的发生密切相关。基因型GA出现频率越高,胃癌发生风险越高(P〈0.05)。胃癌组Hp感染率为59.0%,显著高于对照组的42.O%(P〈0.05)。COX-2基因SNP rs3218625位点,Hp感染阳性及阴性患者携带基因型GA、AA或等位基因A(GA+AA)的频率在胃癌组及对照组中均无显著性差异(P〉0.05)。经Logistic回归分析显示,Hp感染与COX-2基因SNPrs3218625无明显相关性(P〉0.05)。结论张家口坝上地区人群COX-2基因多态性可能会增加当地胃癌的发生风险,其中SNP rs3218625的基因型GA是胃癌发生的危险因素之一,而Hp感染与COX-2基因SNPrs3218625无显著相关性。 Objective To explore the relationship between cyclooxygenase -2 (COW- 2 ) genetic polymorphisms and helicobacter pylori (Hp) infection and the incidence of gastric cancer in Zhangjiakou area. Methods 200 Individuals living in Zhangjiakou area from May in 2015 to February in 2017 were elected and divided into gastric carcinoma group (n = 100) and control group ( n = 100) according to the incidence of gastric cancer. The genetic typing in the two groups was made by Polymerase chain reaction -restriction fragment length polymorphism technique (PCR -RFLP). And the Hp infecftion of the samples in the two groups was detected by Warhin - starry method. Then the relationship between single nucleotide polymorphism (SNP) rs3218625 of COX -2 and Hp infection and the incidence of gastric cancer was analyzed by non - conditional Logistic regression. Results The result of non -conditional Logistic regression showed that COX -2 genetic SNP rs3218625 was closely related to the incidence of gastric cancer (P〈 0. 05 ). And the more the frequency of genotype GA was high, the more the risk of gastric cancer was high (P 〈 0. 05 ). The Hp infection rate in gastric carcinoma group was 57. 0%, which was higher than that in the control group (42. 0% ) (P 〈0. 05). There were no significant difference in the frequency of genotype GA, AA or allele A (GA + AA) , rs3218625 of cox -2, and Hp infection in the two groups (P 〉0. 05 ). Logistic regression showed that Hp infection was not closely related to rs3218625 of COX -2 (P 〉 0. 05). Conclusion The COX - 2 genetic polymorphisms can increase the risk of gastric cancer in Zhangjiakou area. GA of SNP rs3218625 is one of the risk factors of gastric cancer. Hp infecftion is not closely related to rs3218625 of COX -2.
